摘 要:目的研究球形脂联素对高糖刺激下肾小管上皮细胞中线粒体融合蛋白2(mfn2)表达的影响。方法体外常规培养的大鼠肾小管细胞,传代后分五组:正常糖对照组(D-葡萄糖5.5mmol/L)、高糖组(D-葡萄糖30mmol/L),高糖组又分为不同浓度的脂联素处理组(5.0、7.5、10μg/m L脂联素),作用48h后收集细胞。线粒体活性氧荧光探针Mito SOX检测线粒体活性氧的产生,蛋白免疫印迹法检测脂联素(ADPN)及mfn2的表达。结果 1与正常对照组相比,高糖组中脂联素及mfn2表达下调,差异有统计学意义(P<0.05),线粒体活性氧ROS增加,差异有统计学意义(P<0.05);2与高糖组相比,脂联素处理组中,各组mfn2表达增加,差异有统计学意义(P<0.05),且呈浓度依赖性,低剂量脂联素减少了线粒体活性氧ROS含量,差异有统计学意义(P<0.05)。结论高糖下调大鼠小管细胞中脂联素及mfn2的表达,增加了小管细胞线粒体活性氧的产生。体外脂联素干预能够上调mfn2的表达,减少线粒体活性氧ROS产生,发挥抗氧化应激的效应。Objective To research the itffluenees of globular adiponeetin on the expression of mitofusins ( mfn2 ) in renal tubular epithelial cell under high glucose stimulation. Methods Conventionally cultured rat renal tubular cells in vitro, 'after sub- culture were divided into five groups: normal gh, cose control group ( D-glucose 5.5mmool/L) , high glucose group ( D-glucose 30mmol/L) , and the high glucose group was divided into different concentrations of adiponectin treatment group(5.0,7. 5,10μg/mL adiponectin), then cells were collected after 48h. Mitochondrial reactive oxygen species, ROS fluorescent probe MitoSOX was used to detect the production of Mitochondrial ROS, and Western blot was uesd to deteete the expression of adiponectin(ADPN) and mfn2. Results Compared with normal control group, adiponectin and mfn2 of high glucose group downregulated, respectively (P 〈 0. 05 ), Mitochondrial ROS increased, respectively ( P 〈 0. 05 ) ; Compared with high glucose group, adiponectin treatment group, mfn2 expressed increased in each group, respectively ( P 〈 0. 05 ) , and was concentration dependent, low dose adiponeetin reduced mitochondrial ROS content,respectively ( P 〈 0. 05 ). Conelttsion High glucose downregulated the expression of adiponectin and mfn2 in tubular cells in rat, an increased production of tubule ceils mitochondrial ROS. The expression of mfn2 was up-regulated by adiponectin in vitro, and the production of mitochondrial ROS was decreased ,to exert the effect of anti oxidative stress.
