摘  要:目的探讨一种能用于大鼠的快速、易掌握、可重复的测定大鼠尿道压力的实验方法。方法健康雌性大鼠12只,体重250~300g。通过膀胱造瘘管法测定膀胱腹压漏尿点压力(ALPP),采用自制的3F单孔尿道测压管持续水流灌注法,对8只大鼠同步测定尿道压力,自动描记和生成静态尿道压力描记(RUPP)曲线。每只雌性大鼠以随机次序分别测定尿道在3、6、9、12点方向的连续压力。其中6只大鼠以同样方法重复RUPP描记以了解该方法的可重复性。对4个方向的RUPP参数进行统计学分析,并将RUPP曲线特点与文献报道的人类女性RUPP进行比较。结果 ALPP测定值为(68.5±23.8)cmH2O。8只完成RUPP,其中6只完成重复的RUPP测定。功能尿道长度(FUL)和最大尿道闭合压(MUCP)在3、6、9、12点方向的测定值分别为(17.00±1.51)、(18.50±1.77)、(17.38±1.30)、(14.75±2.31)mm和(53.00±17.96)、(46.00±12.60)、(51.50±10.54)、(82.50±28.68)cmH2O,FUL在6点方向最长,MUCP在12点方向最高,与其它3个方向比较,差异有统计学意义(均P〈0.05),FUL和MUCP在3点和9点方向差异无统计学意义。重复描记4个方向的RUPP,无论FUL和MUCP测定值还是RUPP曲线形态与第1次RUPP比较差异均无统计学意义。雌性大鼠RUPP曲线形态与人类女性的RUPP具有高度相似性。结论改良的水灌注法描记雌性大鼠RUPP具有可行性和良好的可重复性。该方法的建立将促进雌性大鼠等小体积实验动物在下尿路功能和病理生理研究中的应用。Objective Rest urethral pressure profile(RUPP)is extensively used to evaluate urethral continence in humans and large-sized animals.But it is still challenging to record RUPP in small-sized animals,like rat.This study explored a novel method for recording RUPP in female rats,with the advantage of less time consuming,easy operation and good repeatability.Methods The abdominal leak point pressure(ALPP)was first determined via a supra-pubic tube in 12 female rats with their weight ranging from 250 to 300g each.The RUPP recording was undertaken using a water-perfusion catheter system with a self-made 3Fsingle-hole urethral piezometric tube.RUPP curves were recorded with the lateral water-perfusion hole facing the3,6,9,or 12o'clock position,respectively,in 8rats at random,and the RUPP recordings were repeated in 6rats.The RUPP parameters recorded at each direction were analyzed and RUPP curve features were compared with those of women reported in the literature.Results The mean ALPP of the rats was(68.5±23.8)cmH2O.RUPP was obtained in eight rats and repeated recording for RUPP was done in six rats.The functional urethral lengths(FULs)at 3,6,9,12o'clock directions were(17.00±1.51),(18.50±1.77),(17.38±1.30),and(14.75±2.31)mm,respectively,and the maximal urethral closure pressures(MUCPs)at 3,6,9,12o'clock directions were(53.00±17.96),(46.00±12.60),(51.50±10.54),and(82.50±28.68)cmH2O,respectively.FUL at 6o'clock and MUCP at 12o'clock were found to be significantly greater than those at other 3directions(P0.05).There were no significant differences in FUL and MUCP between positions at 3and 9o'clock(P0.05).FUL and MUCP at 4directions in repeated RUPP recordings were not significantly different from those in the primary recordings.The RUPP curve shape in female rats was similar to that recorded in women.Conclusion The modified water-perfusion catheter system for RUPP recording is feasible and repeatable in female rats.This method will promote th
