摘  要:为了探讨Nme2(metastasis suppressor non-metastatic 2)基因在男性生殖中的作用,利用GEO(gene expression omnibus,GEO)基因芯片数据库对Nme2在精子发生中的表达情况进行数据挖掘和分析,通过MTT(5-diphenyltetrazolium bromide,MTT)和PI-Hoechst实验构建过氧化氢诱导的GC-1 spg细胞氧化损伤模型,荧光定量PCR技术检测氧化损伤分子OGG1(8-oxoguanine DNA glycosylase),ALDH2(aldehyde dehydrogenase 2family)以及Nme2的表达。发现Nme2在不同类型生精细胞中差异性表达,在畸形精子中的表达显著降低;进一步研究发现Nme2在细胞氧化损伤过程中表达上调。结果表明Nme2与畸形精子症相关,其作用机制可能通过应答氧化损伤信号来实现。In order to study the role of metastasis suppressor non-metastatic 2 (Nine2) gene in male reproduction, the expression of Nme2 in chip databases. Then the oxidation spermatogenesis was analyzed by gene expression omnibus damage model of GC-1 spg cells was induced by hydrogen verified by 5-diphenyhetrazolium bromide (MTY) (GEO) gene peroxide and and PI-Hoechst assay. The expression of oxidative damage molecular markers 8-oxoguanine DNA glycosylase (OGG1), aldehyde dehydrogenase 2 family (ALDH2) and Nme2 were detected. The results showed that Nme2 had different expression profile in different type of spermatogenic cells. In addition, Nme2's expression mRNA level of Nme2 was obviously increased in significantly reduced in teratozoospermia. Furthermore, the response to oxidative damage signal. These results suggest that Nme2 is related to teratozoospermic and its role may be accomplished by response to oxidative damage.
