脓毒血症大鼠模型心脏5-羟色胺与心脏动作电位变化的实验研究

摘　　要：目的探讨脓毒血症大鼠模型心脏5-羟色胺改变与心肌动作电位变化之间的关系。方法利用随机数字表将大鼠分为脓毒血症组和正常对照组,每组各10只,进行脓毒血症模型制作,18 h后进行心脏病理切片、电生理实验、5-羟色胺免疫组化实验并定量分析。结果脓毒血症组心脏线粒体发生改变,其心率[(353.60±11.27)次/min]、心房动作电位时程[(112.16±8.97)ms]均较正常组[心率(306.10±11.21)次/min、心房动作电位时程(86.31±7.49)ms]延长(P<0.01),心肌细胞阳性面积[(19 193.38±588.13)μm2]和阳性细胞数[(18.50±3.37)个/视野]较正常组[(7 755.01±586.16)μm2、(13.30±1.49)个/视野]均明显增加(P<0.05,P<0.01)。结论 5-羟色胺是一种炎性因子,导致心房动作电位时程延长、心率增快,加重心肌损伤,也是导致脓毒血症心功能障碍的众多因素之一。Objective To observe the relation of heart serotonin and myocardial action potential change in the model of sepsis rats. Methods 20 Sprague-Dawley rats were randomly divided into 2 groups, sham operated group（normal group）, controlled operated group（sepsis group）, the model of sepsis rats was made by the way of caecum ligated and punctured, myocardium were taken from all of them 18 h after operation. The activities of pathological section, electrophysiological experiment, immunohistochemical experiment and quantitative analysis of serotonin in myocardial were detected. Results Pathological change of mitochondria in myocardium were observed. The heart rate（353.60± 11.27）bpm, action potentials of heart atrium（112.16±8.97）ms in sepsis group were significantly increased compared with normal group（306.10±11.21）bpm,（86.31±7.49）ms（P〈0.01）, positive area（19 193.38± 588.13）μm2, and the positive cell number（18.50±3.37） every visual field in sepsis group were significantly higher than that of normal group（7 755.01±586.16） μm2,（13.30±1.49） every visual field（P〈0.05, P〈0.01）. Conclusion Serotonin is a kind of inflammatory factor, it can lead to extend atrium action potential and increase heart rate and myocardial injury, it was one of the many factors of cardiac function disorder in sepsis rats.
