摘 要:目的比较使用CFSE作为荧光标记物标记脐血细胞后各组细胞存活率的实验研究。方法使用5μmol/L、10μmol/L、15μmol/L、20μmol/L、25μmol/L、30μmol/L浓度的CFSE工作液对脐血细胞进行标记并计算每组细胞的细胞存活率,进行统计分析。结果 6组不同CFSE终浓度之间的细胞存活率存在显著性差异(P<0.05),且随着CFSE终浓度的增加,细胞存活率有一定程度的下降。不同时间之间的细胞存活率存在显著差异(P<0.01),随着时间的推移,细胞存活率不断下降,且时间差异要比浓度之间的差异显著。结论 CFSE标记的脐血细胞随培养时间延长存活率下降,而浓度对细胞存活率影响较时间小。Objective To compare the survival rate of umbilical cord blood cells using CFSE as fluorescent markers. Methods The final concentration of CFSE cell staining was set to 5 μmol/L, 10 μmol/L, 15 μmol/L, 20 μmol/L, 25 μmol/L, 30 μmol/L respectively, and counted the survival rate of different groups. Results It was significant different in survival rate(P〈0.05) in the 6 groups with different CFSE final concentration. The more final concentration of CFSE operating fluid increased, the lower the survival rate of umbilical cord blood cells was. It was also significant diffenence of the survival rate of the umbilical cord blood cells(P〈0.05). With the time going on, the survival rate of the umbilical cord cells was lower. Time difference was more significant than concentration. Conclusion The survival rate of the umbilical cord cells was as low as the time extension. Concentration has little effect on cell survival rate than time.
