摘 要:目的研究通天草水提物(AEWH)对阿尔茨海默病(AD)小鼠海马形态学和白介素-6(IL-6)水平的影响.方法:雄性小鼠随机分成对照组、模型组、吡拉西坦组(0.62g·kg-1·d-1)、AEWH高剂量组(2.0g·kg-1·d-1)、AEWH低剂量组(0.5g·kg-1·d-1).采用Aβ1-42小鼠双侧脑室注射诱导AD模型小鼠.小鼠给予AEWH灌胃,连续治疗28d后取材,观察小鼠海马镜下形态结构、脾脏和脑的脏器指数.采用双抗体夹心法测定海马IL-6水平.结果:与对照组比较,模型组小鼠神经元数量减少、结构异常,脾脏和脑的脏器指数明显降低(P〈0.05),IL-6水平明显升高(P〈0.05);与模型组比较,AEWH高剂量组海马神经元细胞形态改善明显,脑和脾脏脏器指数明显升高(P〈0.05),IL-6水平明显降低(P〈0.05).结论:AEWH高剂量能维持AD模型小鼠神经元正常形态结构,升高脾脏和脑的脏器指数,降低IL-6水平.ObjectiveEffects of Aqueous Extract from Waternut Herb(AEWH) on morphology and Interleukin 〈app:ds:interleukin〉-6(IL-6) of Hippocampal in Alzheimer's disease (AD) mice. Methods:Male mice were rando mly divided into:control group,model group, Piracetam 〈app:ds:Piracetam%20Tablet〉 group(0.62 g·kg-1·d-1),AEWH high-dose group(2.0g·kg-1·d-1),AEWH low-dose group(0.5g·kg-1·d-1). Aβ1-42 was injected into the bilateral hippocampus of mouse to induce an AD model mice. The mice were killed after 28 days continuous treating. After the treatment all animals were sacrificed,morphology of Hippocampal was observed,calculated the organ coefficients of splenic organ 〈app:ds:splenic%20organ〉 and brain 〈app:ds:brain〉. Biochemical methods were used to determice the content of IL-6 in the Hippocampal. Results:Compared with the control group,the model group lost normal morphological structure of nerve cells and the number of nerve cells was significantly decreased,the organ coefficients of splenic organ 〈app:ds:splenic%20organ〉 and brain 〈app:ds:brain〉 significantly decreased(P〈0.05),IL-6 were significantly increased(P〈0.05).Compared with the model group,the AEWH high-dose group brain maintain normal morphological structure of nerve cells and the number of nerve cells was significantly increased,the organ coefficients of splenic organ and brain 〈app:ds:brain〉 significantly increased(P〈0.05),IL-6 were significantly decreased(P〈0.05).Conclusion:AEWH high-dose group can maintain normal morphological structure of nerve cells of AD model mice,increased organ coefficients of splenic organ 〈app:ds:splenic%20organ〉 and brain 〈app:ds:brain〉,decrease IL-6 responses.
