摘  要:目的:研究黄芪散(HQS)对2型糖尿病(T2DM)模型大鼠血浆磷脂代谢的影响。方法:给予8周龄SD大鼠53.3%高脂饲料13周,建立T2DM模型。在造模的同时灌胃给予黄芪散(HQS),以UPLC/Q-TOF MS为分析手段研究大鼠在2型糖尿病状态以及黄芪散干预下的血浆脂质代谢物的情况。得到大量数据,再通过R软件中BioMark软件包实现对两组样品中特异性的标志物进行筛选。结果:筛选出的标志物经过成分鉴定发现主要为磷脂酰胆碱类(PC)以及其它脂类成分。HQS给药后主要改善了T2DM大鼠血浆中PC类等磷脂成分的代谢紊乱,同时增加血浆中高不饱和PC类磷脂的含量,如PC(40∶8)、PC(40∶7)、PC(40∶4)、PC(42∶9)。结论:黄芪散对T2DM引起的血浆磷脂代谢紊乱有一定的调节改善作用。Objective:This study was report the effect of Huangqi San(HQS) on phospholipids metabolism in Type 2 diabetes mellitus(T2DM). Methods: T2DM model was replicated in 8-week age rats which were administrated with 53.5 % high fat diet for 13 weeks. HQS was simultaneously administrated by intragastric gavage during the modeling. This time we focus on the state of plasma metabolite profiling in nomal body,T2DM model and drug(Huangqi Powder) treatment group using UPLC/Q-TOF MS which was used as analysis tool in this research. Meanwhile it will generate large amount of information-rich MS data which will need chemomettics and lipid databases to help us understand what on earth the unknown compounds are and what roles they play in living bodys. Results:The results show that the pathological markers were mainly phosphatidylcholine (PC). It is mainly improved PC in rat plasma phospholipid metabolism disorder with the normal trend of development after oral administration of HQS. At the same time,increase the content of highly unsaturated PC,such as PC(40 : 8) ,PC(40 :7),PC(40 :4),PC(42 : 9). Conclusion:The experimental results proved that HQS have certain regulation and improvement effects on phospholipid metabolic disorder caused by the body of the T2DM.
