摘 要:目的:建立白花丹中β-谷甾醇含量的HPLC-ELSD快速测定方法,比较野生与家种药材中β-谷甾醇的含量。方法:采用HPLC-ELSD法测定野生与家种白花丹药材根、茎、叶中β-谷甾醇含量。色谱条件为:色谱柱为迪马C_(18)柱(250mm×4.6mm,5μm);流动相为纯甲醇;流速为1.0mL/min;柱温:25℃;以蒸发光散射检测器进行检测,检测参数为漂移管温度65℃,载气(N2)流速为2.5L/min。结果:β-谷甾醇在0.2~3.5μg/mL范围内与峰面积值呈良好线性关系,回归方程为:Y=1.500 3X+5.582 6;β-谷甾醇在叶中含量最高。结论:以β-谷甾醇为主要活性成分时最佳入药部位为叶,白花丹叶为可再生资源,可作为β-谷甾醇的新药用资源植物。Objective:To establish an HPLC-ELSD method of the determination of β-sitosterol in Herba Plumbaginis Zeylanicae; Compare the content of β sitosterol in home-grown and in wild. Methods. The Higher Performance Liquid was used to determine β- sitosterol content in roots, stems and leaves of home-grown and wild Herba Plumbaginis Zeylanicae. Diamondsil C18(250mm × 4.6mm, 5 μm) was used with pure methanol as the mobile phase and the flow rate was 1.0mL/min. The temperature of column was 25 ℃. ELSD nebulization at 65 ℃ and flow rate of nitrogen at 2.5L/min. Results: The linear ranges of β sitosterol were 0.2 3.5μg/mL. The content of β-sitosterol was highest in leaves. Conclusion : Considering β-sitosterol content of Herba Plumbaginis Zeylanicae, The best medicine part was the leaf.
