前列地尔和瓜蒌皮对大鼠心肌缺血再灌注损伤的保护作用研究

摘　　要：目的:通过观察大鼠血清中一氧化氮(NO)和丙二醛(MDA)含量、超氧化物岐化酶(SOD)及肌酸激酶(CK)活性的变化探讨瓜蒌皮和前列地尔预处理对心肌缺血再灌注损伤(myocardial ischemia reperfusion injury,MRI)是否有保护性,并探讨其可能的作用机制。方法:建立SD大鼠的MRI模型,通过Tunel法检测组织样本的细胞凋亡情况,并收集各实验组MRI后的心脏血液,然后分离血清,使用试剂盒检测各组血清中NO和MDA含量、SOD及CK活性进行研究。结果:瓜蒌皮和前列地尔半剂量联合组的凋亡细胞数、MDA含量、CK活性较其他各实验组均明显降低,SOD活性、NO含量升高,且有统计学意义(P<0.05);瓜蒌皮单用药组也表现为凋亡细胞数、MDA含量、CK活性的表达量降低,SOD活性、NO含量升高,但程度不如两组联合用药组。结论:在MRI中,瓜蒌皮可起到一定的保护作用,并且瓜蒌皮与前列地尔联合应用更有利于对心肌细胞的保护^([1])。Objective：To investigate the protective effects of trichosanchis and alprostadil on myocardial ischemia-reperfusion injury by observing the changes of NO,MDA,SOD and CK in the rat serum.Method：The apoptosis was evaluated with Tunel and the expression of NO,MDA,SOD and CK in serum was detected in a rat model of myocardial ischemia-reperfusion injury.Result：When compared with other groups,the therapeutic group of pericarpium trichosanthis combined with alprostadil showed reduced the number of apoptotic cells,the content of MDA and the activity of CK,but increased the activity of SOD and the content of NO（P〈0.05）.In addition,the therapeutic group of pericarpium trichosanthis has similar protective effects,but was much weaker than the combined group.Conclusion：Thichosanthes may provide the protective effects against myocardial ischemia-eperfusion injury,partially combined with alprostadil.
