新西兰兔心肌缺血后血清H-FABP含量变化及桂枝汤复合方剂的干预研究

摘　　要：目的:研究新西兰兔心肌缺血后血清中心型脂肪酸结合蛋白(heart-type fatty acid-binding protein,H-FABP)的动态变化,以及中药桂枝汤复合方剂的干预作用。方法:将24只兔随机分为正常组、模型组、药物组,每组8只。药物组用桂枝汤复合方剂(6g·kg^(-1)·d^(-1))灌胃14d,正常组和模型组给予等量生理盐水灌胃14d。制作急性心肌缺血模型(药物组和模型组),正常组只穿线不结扎。采用结扎左冠状动脉前降支的方法复制心肌缺血模型。ELISA法检测血清H-FABP的含量,彩色多普勒超声心动图观察心脏射血分数(EF)。结果:与正常组相比,模型组结扎后30min H-FABP浓度开始升高,4h达高峰,6h开始下降(P<0.05);与模型组相比,药物组结扎后各时间点H-FABP含量均降低,结扎后6h心脏射血分数增高(P<0.05)。结论:血清H-FABP含量随心肌缺血时间有动态变化,中药桂枝汤复合方剂的预防应用可以降低血清H-FABP含量,改善心功能状态。Objective：To observe dynamic changes of serum Heart-type fatty acid-binding protein（H-FABP）in myocardial ischemia New Zealand rabbit and intervention effect of Guizhi soup.Method：Twenty-four rabbits were randomly divided into three group：nomal group,model group and drug group.The myocardial ischemia model was created by left anterior descending coronary artery ligation.ELISA was used to detect serum content of H-FABP,Color Doppler Echocardiography was performed to measure ejection fraction（EF）.Result：H-FABP concentrations began to rise at 30 min reached a peak at 4h,and then gradually decreased at 6h（P〈0.05）after myocardial ischemia.Compared with the model group,H-FABP content is lower but EF is higher in drug group（P〈0.05）.Conclusion：Serum H-FABP content dynamically changes after myocardial ischemia and treatment with guizhi soup can reduce serum H-FABP content and improve heart function.
