摘 要:为了解鸡干扰素刺激基因(ISG)在鸡传染性支气管炎病毒(IBV)感染雏鸡气管黏膜中的表达情况,采用实时荧光定量PCR检测IBV M41株感染SPF鸡后不同时相气管黏膜中β干扰素(IFN-β)和干扰素刺激基因mRNA转录水平和IBV病毒载量变化。结果显示,鸡气管黏膜中IFN-β和干扰素刺激基因Mx、PKR、OAS、Viperin、IFITM3和IFIT5mRNA转录水平在感染后(DPI)第1~14天显著(P〈0.05)或极显著(P〈0.01)上调,且均在5DPI达到峰值;鸡气管黏膜中IBV病毒载量在1DPI急剧升高,在5DPI达到峰值,14DPI病毒载量急剧下降。结果表明,IFN-β、干扰素刺激基因Mx、PKR、OAS、Viperin、IFITM3和IFIT5mRNA转录水平与IBV病毒载量变化趋势一致,提示IFN-β、干扰素刺激基因Mx、PKR、OAS、Viperin、IFITM3和IFIT5在抵御IBV入侵、抑制IBV复制、增殖以及病毒清除中发挥重要作用。试验结果丰富了宿主抗IBV感染的固有免疫应答机制,为IBV感染的预防和控制提供了理论依据。In order to understand the expression of interferon stimulated genes(ISG)in trachea mucosa of chickens infected with infectious bronchitis virus(IBV),the mRNA transcription levels of IFN-β,interferon stimulated genes Mx,PKR,OAS,Viperin,IFITM3 and IFIT5,as well as IBV viral load in tracheae of chickens at different time points after infection with IBV M41 strain were detected by real-time PCR method in the present study.The results showed that the mRNA transcription levels of IFN-βand interferon stimulated genes Mx,PKR,OAS,Viperin,IFITM3 and IFIT5were significant(P0.05)or highly significant(P0.01)up-regulation during 1DPI to 14 DPI,and reached the peak also at 5DPI.The IBV viral load rose rapidly at 1day post infection(DPI),and reached the peak at 5DPI,then decreased rapidly at 14 DPI.The present results revealed that change trend of the mRNA transcription levels of ISG was consistent with that of IBV viral load.FN-βand interferon stimulated genes Mx,PKR,OAS,Viperin,IFITM3 and IFIT5played an critical role in resisting IBV invasion,suppressing the replication and proliferation of IBV as well as viral clearance.This study enriched the host innate immune response mechanism against IBV infection and provided theoretical basis for the prevention and control of IBV infection.
