荧光假单胞菌疫苗对异育银鲫的免疫保护效应

摘　　要：为探讨荧光假单胞菌疫苗对异育银鲫的免疫保护作用,为其在赤皮病防治上的应用提供理论依据,以福尔马林灭活的荧光假单胞菌(Pseudamonas fluoroscens)全菌疫苗、菌体外膜蛋白(OMP)疫苗作为免疫原,以平均体重100.0g±15.0g的健康异育银鲫(Carassius auratus gibelio)为试验对象,通过腹腔注射免疫原(浓度分别为1.0×108/mL、0.2mg/mL)免疫,剂量为0.2mL/尾,对照组注射等量灭菌生理盐水。分别在免疫注射1、2、4、7、14、21、28、35d后对试验鱼尾静脉采血,进行外周血细胞计数、吞噬活性和抗体效价的测定。结果2种免疫原均可诱导异育银鲫外周血液红细胞、白细胞数量增加,吞噬细胞的吞噬活性和血清抗体效价均升高。免疫注射35d后活菌攻毒,福尔马林灭活荧光假单胞菌苗(F-PF)组的免疫保护率为61.1%,OMP组的免疫保护率为72.2%,OMP组的免疫保护效果优于F-PF组。证明2种免疫原均通过促进异育银鲫血细胞增殖,产生特异性抗体的方式提高机体的免疫保护力。To investigate the immune protection effects of Pseudamonas fluoroscens vaccens on Carassius auratus gibelio and provide important scientific basis on prevention and treatment of red skin disease.Formalin-killed Pseudamonasfluoroscenscells（F-PF）and outer membrane protein（OMP）were used as immunogen to immunize healthy Carassius auratus gibelio with an average body weight of 100.0g±15.0g.Each individual was intraperitoneally injected with 0.2mL 1.0×108 mL formalin-killed Pseudamonas fluoroscens in F-PF group and 0.2mL 0.2mg/mL outer membrane protein in OMP group,and with 0.2mL6.5mg/mL sterile saline in control group,respectively.Then,approximate 2mL of blood was taken from each fish caudal vein on day 1,2,4,7,14,21,28 and 35post injection to study the changes of immune parameters,such as haematocyte number,phagocytic activity of phagocytes,serum antibody titer.The results showed that increasing number of erythrocytes and leucocytes in peripheral blood and phagocytic activity of phagocytes of immunized Carassius auratus gibelio were made by two vaccines;The result of challenge with live Pseudamonas fluoroscens showed that Carassius auratus gibelio immunized with two vaccines had good relative percent survival（RPS）.OMP group had the higher RPS,which was 72.2%.F-PF group had the RPS with 61.1%.All the results stated above suggested that two vaccines could enhance the immunity of Carassius auratus gibelio,such as haematocyte number,serum antibody titer,against bacterial pathogen infection.
