摘　　要：镰刀菌不仅能侵染小麦、大麦、玉米等粮食作物,造成作物产量严重降低,谷物中存留的镰刀菌毒素还能引发人、畜中毒,严重威胁人、畜健康。本研究以镰刀菌细胞壁蛋白特异单链抗体CWP2为靶标,淘选噬菌体随机12肽库,筛选获得两种与抗体CWP2特异结合的短肽,多肽1(YLNPPCDLYACA)和多肽2(YQSVFFTDPHEF)。竞争性酶联免疫吸附实验证实,多肽1更好的模拟了镰刀菌细胞壁蛋白的抗原表位。这是首次对镰刀菌细胞壁蛋白模拟肽的报道,为探寻CWP2所结合抗原表位的编码基因,进一步阐明CWP2介导的抗镰刀菌分子机理提供了信息和材料。同时,获得的模拟肽可直接用于家畜疫苗以及作为粮食安全检测标准品。Fusarium can infect not only wheat, barley, corn and other food crops, resulting in severely reduced crop yields, but even the toxin in stored grain produced by Fusarium can make the human and livestock poisoning, which gravely threaten the health of human and animal. In this study, the single-chain （scFv） antibody CWP2 against the cell wall proteins of Fus ariurn was used as target protein to pan oligopeptides from Ph.D.-12 phage display peptide library, and two polypeptides YLNPPCDLYACA （peptide 1） and YQSVFFTDPHEF （peptide 2） were obtained which can specifically combine with CWP2. And the result of competitive enzyme-linked immunosorbent assay （ELISA） confirms, the peptide 1 has better simution with epitope of the cell wall proteins of Fusarium. This is the first report about cell wall protein mimotopes of Fusarium, which will lay the foundationwill provide the information and material for exploring the coding gene for antigen recognized by CWP2 antibody and further clarifying the mechanism of CWP2 antibody mediated resistance against Fusarium. Meanwhile, the mimic peptide we obtained has potential applications can directly used in livestock vaccineand new standard sample for food safety test.
