摘 要:目的 建立了微流控芯片非接触电导检测法测定三颗针与盐酸小檗碱片中小檗碱的分析方法。方法 选择5 mmol/L乙二胺+15 mmol/L H_3BO_3作缓冲溶液;加入0.4 mmol/Lβ-环糊精(β-CD)添加剂;分离电压2.50 kV;进样时间10 s;结果 盐酸小檗碱的线性范围为10-4.0×10-2μg/mL(r=0.997),检出限为10μg/mL(S/N=3),三颗针中小檗碱平均含量为0.14%,盐酸小檗碱片中小檗碱平均含量为91.6 mg/片,其平均标准偏差(RSD)分别为1.3%(n=6)、1.4%(n=6)。平均加标回收率分别为103%和98.9%。结论 该方法快速、简单、高效,可为三颗针及盐酸小檗碱片提供新的分析方法。Objective To establish a new method for the determination of Berberine in Barberry Root and Berberine hydrochloride Tablets by mierofluidic chip with contactless conductivity detection. Methods 5 mmol/L ethylene diamine + 15 mmol/L boric acid + 0.4 mmol/L β-CD were selected as buffer solution, 2.50 kV was chosen as separation voltage with injecting time of 10 s. Results The linear range of berberine ranged from 10 to 4.0×10^2 μg/mL (r = 0.997) with the detection limit of 10 μg/mL (S/N = 3), the average recovery was 103% and 98.9% respectively. The average concentration of berberine in barberry root was 0.14% (RSD = 1.3%, n = 6) and the average content of berberine in berberine hydrochloride tablet was 91.6 mg (RSD = 1.4%, n = 6). Conclusion The method is simple, rapid and efficient, which can provide a new analysis method for Berberine and Barberry root.
