摘  要:建立知母中知母皂苷BⅡ的含量测定方法。采用TLC法鉴定了知母中知母皂苷BⅡ;用高效液相色谱-蒸发光散射检测(HPLC-ELSD)方法测定知母中知母皂苷BⅡ的含量。色谱柱:Alltima C18(4.6mm×250 mm,5μm);流动相:乙腈-水(25∶75);流速:1.0m L/min;ELSD检测器漂移管温度:100℃;载气流速:3.0m L/min。知母皂苷BⅡ在2.0μg^80.0μg范围内线性关系良好,R为0.9996。平均回收率为99.5%,RSD为1.89%。本方法操作简单、快速灵敏,可作为知母中知母皂苷BⅡ含量的测定方法。To develop a method for the determination of timosaponin BⅡ in Rhizoma Anemarrhena. By thin layer chromatography timosaponin BⅡ in Rhizoma Anemarrhena was determined,and the content of timosaponin BⅡ in Rhizoma Anemarrhena was derermined by HPLC- ELSD. It was performed on an Alltima C18( 250 mm × 4. 6mm,5μm) column using acetonitrile- water( 25∶ 75) as the mobile phase at the flow rate of 1. 0m L / min. The temperature of drift tube was 100℃,and the flow volume of gas was 3. 0m L / min. Good linearity was found between 2. 0μg ~ 80. 0μg and the correlation coefficient was higher than 0. 9996. The average recovery rate was 99. 5%with relative standard deviation( RSD) values being 1. 89%. The developed method was simple,rapid and sensitive,and can be used for the content determination of timosaponin BⅡ.
