摘  要:目的:观察滋阴清热药对狼疮鼠骨髓EMP及血清抗ds-DNA抗体的影响.方法:以滋阴清热方干预狼疮鼠,免疫组化方法观察EMP阳性细胞在骨髓的组织形态,定量PCR检测EMP基因表达,同时检测血清抗ds-DNA抗体浓度和尿蛋白.结果:狼疮模型鼠骨髓EMP阳性细胞比正常对照组密集,基因表达增高,血清抗ds-DNA抗体浓度和尿蛋白也更高;滋阴清热药狼疮模型鼠组与生理盐水狼疮模型鼠组EMP的基因表达组间比较明显下降,差异有统计学意义(P<0.05);血清抗ds-DNA抗体浓度和尿蛋白也明显下降,差异有统计学意义(P<0.05).结论:EMP可能是滋阴清热药治疗SLE新的作用靶点.AIM: To observe the effect on erythroblast macrophage protein( EMP) and anti ds-DNA antibody in bone marrow tissue of lupus mice with Nourishing Yin Clearing Heat Herbs.METHODS: Lupus mice were treated with Nourishing Yin Clearing Heat herbs. The histomorphology of EMP positive cells in bone marrow were observed by immunohistochemistry,EMP gene expression were detected by quantitative PCR,and the concentration of serum anti ds-DNA antibody and Proteinuria was detected respectively. RESULTS: Compared with normal control group,the lupus mice had more intensive of EMP positive cells and higher EMP gene expression in bone marrow,also had higher concentration of serum anti ds-DNA antibody and Proteinuria. Compared with lupus mice control group,the intensive of EMP positive cells and EMP gene expression of the Nourishing Yin Clearing Heat Herbs lupus mice group was decreased,and concentration of serum anti ds-DNA antibody and Proteinuria was also decreased.CONCLUSION: EMP may be a new target of treatment for systemic lupus erythematosus( SLE) by Nourishing Yin and Clearing Heat Herbs.
