摘  要:实验教学是高校分子生物学专业人才培养过程中不可或缺的关键环节,开发综合性和设计性实验项目可提高分子生物学实验教学的质量。CRISPR/Cas9是近年快速发展起来的基因组编辑技术,目前已成功应用于诸多物种,包括猪、兔和农作物等。以CRISPR/Cas9基因组编辑技术为核心,本实验室开设了一门名为"基因组定点编辑技术在动植物中的应用"创新性实验教学课程。本文详细阐述了实施该课程的教学目标、重点与难点、实验原理、实验步骤及结果等。实践表明,此实验教学项目可巧妙地将生物化学、分子细胞生物学、基因工程技术、生物信息学等多学科知识点有机整合。采用此策略,不仅可巩固与加强学生从分子生物学理论课所学的基础知识,同时有利于提高其综合实验设计能力与培养创新精神。该创新性实验课程内容难度适中,包含有验证性和探索性实验,可在开设分子生物学专业的高校推广与试行。Experimental teaching plays an important role in cultivating the molecular biology talents in higher education. To improve the quality of experimental teaching of molecular biology, the comprehensive design of experiments must be developed. CRISPR/Cas9 is a genome editing technology which developed rapidly and has been successfully applied in many organisms, including pig, sheep, rabbit and crop plants. Based on the CRISPR/Cas9 technology, an innovative experimental teaching course named "Targeted genome editing technology and its application in animals and plants" has been constructed. In this study, the teaching objective, focus and difficulty, experimental principles, experimental procedures and results of this course are detailed. In fact, the innovative experimental teaching course is able to flexible integrated knowledge of biochemistry, molecular and cellular biology, technique for gene engineering and bioinformatics and so on. It indicates that applying this approach is not only beneficial to consolidate and strengthen students' knowledge which learning from course of molecular biology on basic theory, but also help students improve their ability about comprehensive designing experiments and the cultivation of the society required innovation. The content of integrated curriculum is not difficult, containing replication and exploratory experiments, which can be widely applied in the other colleges and universities.
