摘  要:目的通过细胞模型和人群研究,探讨血清δ-氨基乙酰丙酸脱水酶(ALAD)含量在铅暴露人群中的分布情况,为铅中毒早预防、早诊断提供依据。方法 Real-time RT-PCR检测铅处理HEK293细胞模型中ALAD变化情况;ELISA方法检测铅暴露人群和非铅暴露人群中血清ALAD、游离红细胞原卟啉(FEP)和平均红细胞血红蛋白量(MCH),分析其在铅中毒过程中的变化情况。结果 HEK293细胞铅处理后ALAD基因mRNA表达水平显著下降(P<0.001);人群中ALAD和MCH含量与血铅浓度呈负相关关系(P<0.001),ALAD和FEP呈负相关关系(P<0.001),且ALAD含量在血铅浓度>300μg/L及≤300μg/L时差异有统计学意义。结论 ALAD作为铅中毒的一个敏感指标,可为职业性铅中毒早期预防和诊断提供某些依据,为铅中毒新标准制定提供一定参考。Objective To explore the level and distribution of serum ALAD in lead exposed population by cell experiment and epidemiological investigation to provide some data for early prevention, diagnosis, and treatment of lead poisoning.Methods Real-time RT-PCR was used to detect the ALAD in lead exposed HEK293 cell model,ELISA was used to detect the serum ALAD、FEP and MCH. Results It was showed that the expression level of ALAD was significantly decreased after treatment with Pb in HEK293 cells model( P 〈0. 001); there was a negative correlation between blood lead and serum ALAD or MCH in lead exposed population( P〈0. 001),ALAD was negatively correlated with FEP( P〈 0. 001); meanwhile,a significant difference of serum ALAD was found between the people whose blood lead levels lower or higher than 300 μg / L. Conclusion The results suggested that ALAD,as a sensitive marker of lead poisoning,could offer information for early prevention and early diagnosis of lead poisoning; it might also provide some reference for the early diagnosis of lead poisoning.
