摘 要:目的探讨钒暴露与热休克蛋白(Heat Shock Protein,HSP)70-2基因多态性对作业工人神经行为功能的影响。方法采用整群抽样抽取某钒制品厂和某化工厂符合条件的各128例工人作为接钒组和对照组,用神经行为核心测验组合(NCTB)测试研究对象的神经行为功能;通过聚合酶链反应-限制性片段长度多态性(PCR-RFLP)的方法来检测作业工人hsp70-2(rs1061581)位点的单核苷酸多态性。结果 (1)在hsp70-2位点上,接钒组和对照组在神经行为功能上的差异主要表现为:在A/G基因型组中,接钒组在错误次数、视觉保留、数字跨度(倒序)、提转捷度(非利手)、目标追踪(正确点数、总打点数)6项上得分更低且差异有统计学意义(P<0.05);而在A/A基因型组中,接钒组仅在数字跨度(倒序)上得分更低且差异有统计学意义(P<0.05);在G/G基因型组中,接钒组仅在错误次数、数字译码两项上得分更低且有统计学意义(P<0.05)。采用主成分分析法构建了神经行为综合指数(Neurobehavioral Index,NBI),在A/G基因型组中,接钒组的NBI得分更低且差异有统计学意义(P<0.05)。(2)接钒组在包括提转捷度(非利手),目标追踪(正确点数,总打点数),数字跨度(顺序)以及NBI得分均以带有A/G基因型的一组得分最低,但各基因型间差异均无统计学意义(P>0.05),对照组未发现此规律。(3)析因分析未发现钒暴露与hsp70-2多态性存在能影响调查对象神经行为功能的交互作用。结论在神经行为表现上,含有A/G基因型的个体在神经行为表现上更容易受到钒暴露的影响,尚未观察到hsp70-2与钒暴露间存在交互作用。Objective To study the effects of hsp70- 2 polymorphism and vanadium exposure on human neurobehavioral function. Methods The demographic information and occupational exposure data were collected using questionnaires. Neurobehavioral Core Test Battery( NCTB) WHO recommended was applied to evaluate the neurobehavioral functions of 128 vanadium- exposed workers and 128 non- exposed workers. After that,Neurobehavioral Index( NBI) were formulated by Principle Component Analysis. And then,the hsp70- 2 genotypes at the rs1061581 site were analyzed by polymerase chain reaction- restriction fragment length polymorphism( PCR- RFLP). The interaction between vanadium-exposure and hsp70- 2 polymorphism was analyzed using factorial analysis. Results The scores of vanadium- exposed workers with A / G genotype were significantly lower than ones of the control in error dot score,visual retention,digital span( backward),Santa Ana dexterity( Non- preferred hand) and pursuit aiming( right,total points)( P〈 0. 05). The scores of vanadium- exposed workers with A / A genotype were significantly lower than ones of the control just in digital span( backward)( P 〈0. 05). The scores of vanadium- exposed workers with G/G genotype were significantly decreased in error dot score and digital symbol( P 〈0. 05). Furthermore,the vanadium- exposed group with A / G genotype performed worse in NBI compared with the control group( P 〈0. 05). A / G genotype of the vanadium- exposed group had a worse performance in Santa Ana dexterity( non- preferred hand),pursuit aiming( right,total points),digital span( forward) and NBI compared with other genotypes,however,there were no significantly statistical difference among different genotypes( P〉 0. 05). Vanadium- exposure and hsp70- 2 interaction was not observed( P〈 0. 05). Conclusion Compared with the other genotypes,the vanadium-exposed workers with hsp70- 2 A / G genotype performed worse on memory and locomotor activity. No intera
