摘  要:目的筛选肾综合征出血热滴鼻疫苗壳聚糖载体,为进一步研究其滴鼻或气雾疫苗提供依据。方法 48只昆明鼠按完全随机分组的方法分为4组,每组12只,采用3种不同粘度的壳聚糖作为载体,制成终浓度为0.1%壳聚糖的病毒液进行鼻腔接种,采用ELISA方法检测肾综合征出血热特异性Ig G抗体。结果疫苗接种后,小鼠能够对各剂量组产生免疫反应,Ⅱ、Ⅲ组能诱导较高、较快血清抗体滴度的增加,免疫后3周,血清抗体滴度维持在较高的水平,但各个实验组间无统计学差异(P>0.05)。结论本实验得出壳聚糖能够增强疫苗的抗体反应,Ⅱ、Ⅲ组效果更佳。Objective An animal experiment was established to evaluate the efficacy of the hemorrhagic fever with renal syndrome intranasal vaccine using chitosan as an adjuvant,preparing for the further study of hemorrhagic fever with renal syndrome intranasal vaccine. Methods 48 mice were randomly divided into 4 groups and the number of every group was equal. By using three different viscosity of chitosan as a carrier,animals were inoculated with intranasal vaccine with 0. 1%chitosan. Blood samples were collected from tail veins every week for detecting the specific Ig G by indirect ELISA. Results Ig G were induced in the mice inoculated with the hemorrhagic fever with renal syndrome intranasal vaccine and were maintained at a high level 3 weeks after inoculation at Ⅱ,Ⅲ groups. There was no significant difference in the antibody titers between different groups. Conclusion The hemorrhagic fever with renal syndrome intranasal vaccine using chitosan as immune adjuvant approach could enhance antibody response,Ⅱ and Ⅲ groups had better effects.
