摘  要:目的:本课题在成功建立AD大鼠模型基础上,观察固本健脑法治疗AD的作用及机制。方法:12月龄衰老大鼠行脑立体定位手术于双侧海马注射Aβ_(1-42),制备复合AD大鼠模型,并用固本健脑液灌胃干预,以安理中组为对照。4周后,高尔基染色法观察大鼠海马区神经元树突棘形态及密度;Western Blot法检测大鼠海马区Caveolin-1、P-tau(Thr231、Ser396位点)蛋白表达改变。结果:高尔基染色法结果显示:固本健脑液高、低剂量组神经元细胞及树突棘形态明显改善,数目及密度显著增加(P<0.01)。Western Blot法检测结果显示:固本健脑液高、低剂量组可使Caveolin-1的表达不同程度的升高,而tau磷酸化水平下降,差异有显著性(P<0.01)。结论:固本健脑法能显著改善AD大鼠海马神经元树突棘形态及密度,显著提高AD模型大鼠海马区Caveolin-1的表达,抑制tau蛋白过度磷酸化。Based on the successful establishment of Alzheimer's disease(AD) rat model,the effects and mechanisms of Gu-Ben Jian-Nao(GBJN) recipe in AD treatment were investigated.The compound AD rat model was prepared with the operation of bilateral hippocampal injection of Aβ_(1-42) on 12-month aged rats.And then,intragastric administration of GBJN liquid was given to the rat model.Aricept was used as control.After 4 weeks,Golgi staining was used to observe the morphology and density of dendritic spines of neurons in the hippocampus of rats.Western blot was used in the detection of changes of Caveolin-1,P-tau(Thr231,Ser396 sites) protein expressions in the hippocampus of rat.The results of Golgi staining showed that both the high and low dose GBJN liquid group can obviously improve the morphology as well as significantly increase the amount and density of dendritic spines of neurons(P〈0.01).Western blot results showed that both the high and low dose GBJN liquid group can increase the Caveolin-1 expression at different degrees,but the tau phosphorylation level decreased,with significant differences(P〈0.01).It was concluded that GBJN recipe can obviously improve the morphology and density of dendritic spines of neurons in the hippocampus of AD rats,significantly increase the Caveolin-1expression in the hippocampus of AD rats,and inhibit the excessive phosphorylated tau protein.
