摘 要:目的:利用超高效液相串联四级杆飞行时间质谱(UPLC-Q-TOF/MS)法分析车前子及其盐炙品的成分,比较盐炙前后药物化学成分种类和含量的变化,探究车前子炮制前后药效变化的物质基础。方法:ACQUITY UPLC BEH C_(18)色谱柱;流动相:0.1%乙酸水-甲醇体系梯度洗脱;ESI离子源;负离子模式;基于Markerlynx分析软件,利用主成分分析法(PCA)和正交偏最小二乘判别法(OPLS-DA)分析车前子盐炙前后指纹图谱的差异。结果:盐炙品中京尼平苷酸、毛蕊花糖苷、异毛蕊花糖苷的含量相对生品高。京尼平苷酸和毛蕊花糖苷可作为化学标记物区分生品与盐炙品。结论:基于UPLC-Q-TOF/MS法建立了车前子炮制前后的化学成分分析方法。车前子盐炙后有效成分含量增加。本实验的研究成果对于研究车前子盐炙的炮制原理具有重要意义,同时也为车前子盐炙品药效物质基础的阐明提供了重要依据。This study was aimed to investigate the changes of chemical compositions and contents of crude and salt-processed Plantago asiatica L.by the ultra-performance liquid chromatography coupled with quadrupoletime-of-flight mass spectrometry(UPLC-Q-TOF/MS) in order to explore the material basis of effect changes before and after the processing.The chromatography separation was performed on the ACQUITY UPLC BEH C_(18)column with gradient elution of 0.1%water(acetic acid)-methanol.The mass spectrometry was equipped with electrospray ionization(ESI) source.The detection was under the negative ion mode.Based on the Markerlynx software,principal component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLSDA) were used in the analysis of the differences on fingerprint before and after the salt-processing.The results showed that in the salt-processed P.asiatica L.,the contents of geniposidic acid,acteoside and isoacteoside were higher than the crude material.Geniposidic acid and acteoside can be used as the chemical markers for the differentiation between the crude and salt-processed materials.It was concluded that the chemical composition analysis method based on UPLC-Q-TOF/MS was established on the study of crude and processed P.asiatica L.After salt-processing,the effective contents in P.asiatica L.were increased.The experiment results played an important role in the processing principle study of salt-processed P.asiatica L.It also provided important evidences for the interpretation of effective material basis on salt-processed P.asiatica L.
