摘  要:目的:研究环磷酰胺(CP)小鼠少精子模型的构建方法,以及评价枸杞煎提液对少精子症的治疗作用。方法小鼠分别以60、80、100mg/kg CP腹腔注射,qid,连续5d,测定精子相对数和睾丸组织切片,得出最适CP剂量;再以高低剂量(20g/kg、10g/kg)枸杞煎提液给最适CP剂量构建的少精子小鼠灌胃14d,测定精子相对数、睾丸组织切片和流式细胞仪检测生精细胞倍性。结果3种剂量CP处理组精子相对计数、生精小管面积、直径、生精上皮细胞层数(CMSE)均显著低于空白对照组和生理盐水组(P<0.001);枸杞高低剂量组精子相对计数、生精小管面积、直径、CMSE显著高于阳性对照组(P<0.001);枸杞高剂量组与阳性对照组比在单倍体细胞(1C细胞)比例上显著降低,四倍体细胞(4C细胞)比例上显著升高(P<0.05),枸杞低剂量组与阳性对照组比在二倍体细胞(2C细胞)比例上显著升高(P<0.05)。结论60mg/kg CP为构建小鼠少精子模型的适宜剂量;枸杞煎提液对少精子症有一定治疗效果,可能与促进4C细胞增殖有关。Objective To establish Cyclophosphamide(CP)-induced oligopermia mouse model and evaluate the curative effect of Matrimony vine on oligospermia.Methods Male mice were intraperitoneally injected with CP of 60, 80, 100mg/kg-1/d-1 for 5 day, then they were comparatively analyzed by means of epididymal sperm count, paraffin sections of testis for a suitable CP dose. The established oligospermia mouse models which were induced by the most suitable CP dose were intragastrically administrated with high and low doses of Matrimony vine (20g/kg, 10g/kg) for 14d, and then epididymal sperm count, paraffin sections of testis and Spermatogenic cells test were analyzed respectively.Results Globus minor relative sperm count, area and diameter of seminiferous tubules, count measure spermatogenic epithelium (CMSE) in three doses of CP group were all significantly lower than those in the blank control group and saline group (P 〈0.001); In high and low doses of Matrimony vine groups, area and Globus minor relative sperm count and diameter of seminiferous tubules, CMSE were all significantly higher than those of the positive control group (P〈0.001); the wolfberry high dose group was lower than the positive control group in the proportion of 1C cells and higher in the proportion of 4C cells (P〈0.05); The wolfberry low dose group was higher than the positive control group at the ratio of 2C cells (P〈0.05).Conclusion The dose of 60mg/kg is the best for establishment of oligopermia mouse model. Matrimony vine has therapeutic effects on model mice which may be associated with an increase in the number of tetraploid cells.
