摘  要:为探究运动发酵单胞菌(Zymomonas mobilis)ZM401的絮凝机理,研究了20种理化因素对ZM401絮凝的影响。结果表明:ZM401的絮凝物质在酸性环境中易降解而在碱性环境中稳定,并且对温度不敏感。金属阳离子的添加对ZM401的絮凝无影响,仅镁离子浓度的增加会使絮凝率提高2.9%。单糖的添加对絮凝没有影响,蔗糖和纤维二糖的增加会提升絮凝率。有机溶剂的添加会增加ZM401的絮凝率,其中10%的乙醇添加使絮凝率提升了2.6%,说明絮凝物质间的结合受疏水作用影响。ZM401的絮凝率在α-淀粉酶、壳聚糖酶、脂肪酶、蛋白酶K添加时无变化,但其对纤维素酶非常敏感,0.15FPU/mL的纤维素酶即可使絮凝颗粒不可逆地解体。上述实验结果证明了运动发酵单胞菌的絮凝物质是纤维素类物质,其相互作用为分子间物理搭桥。The effect of 20 kinds of physical and chemical factors on the flocculation of Zymomonas mobilis ZM401 has been studied to reveal the underlying mechanism of flocculation.The results show that the flocculation material of ZM401 can be easily degraded in acid,but is stable in alkaline environment.Moreover,the flocculation is not sensitive to temperature.The addition of metal cations has no obvious effect on the flocculation,only with the incline of Mg^2+,the flocculation efficiency has a slightly increase by 2.9%.Monosaccharide has no effect on the flocculation,while sucrose and cellobiose have postive effect on the flocculation efficiency.The addition of organic solvents increases the flocculation,and 10% ethanol improves the flocculation efficiency by 2.6%.The addition of theα-amylase,chitosanase,lipase and protease K almost has no influence on the flocculation.But flocculation is very sensitive to cellulase,which with 0.15FPU/mL causes irreversible collapse of flocculation.It is proved that the material related to flocculation of Z.mobilis is cellulose,by which cell adhere to each other.
