摘  要:目的探讨自拟控颤汤对帕金森病大鼠黑质神经元保护作用的机制。方法利用立体定向仪向SD大鼠黑质内注射6-羟基多巴胺成功复制帕金森病大鼠模型,大鼠共分为四组:模型组(n=10)、多巴丝肼治疗组(西药组,n=10)和自拟控颤汤治疗组(中药组,n=10)以及对照组(n=8,未造模),给药后第14天和第28天进行旋转行为学测试,术后4周取大鼠的术侧中脑黑质利用免疫组化检测酪氨酸羟化酶(TH)免疫阳性细胞的表达,并进行统计学分析,判断疗效。结果中药组和西药组帕金森病大鼠治疗后第14天和第28天均较治疗前旋平均转速降低,差异有统计学意义(P<0.05),而模型组治疗前后则无变化;与对照组细胞数(112.34±14.46)比较,模型组TH阳性细胞数(49.83±11.74)显著减少,差异有统计学意义(P<0.05);与模型组比较,西药组和中药组TH阳性细胞数(86.29±12.28、67.45±13.80)显著增加,差异有统计学意义(P<0.05)。结论自拟控颤汤能改善帕金森病大鼠旋转行为,并参与大鼠黑质多巴胺能神经元保护作用,通过实验研究进一步验证了自拟控颤汤对帕金森病的治疗效果。Objective To explore the protection mechanism of self-made control tremor decoction on neurons in the substantia nigra of rats with Parkinson disease. Methods 6-hydroxy dopamine was injected into the substantia nigra of SD rats by using of stereotactic instrument to make rats model of Parkinson disease. The rats were divided into four groups :model group( n = 10), Benserazide treatment group (Western medicine group, n = 10)and self-made control tremor decoction treatment group (Chinese medicine group, n = 10) and control group( n = 8, without modeling). The rotation behavior test was carried out after the using of drug for fourteenth days and twenty-eighth days. The expression of tyrosine hydroxylase(TH)positive cells in Substantia nigra of midbrain was detected by immunohistochemistry 4 weeks after operation. And the statistical analysis was performed to determine the effect of the treatment. Results The rotation numbers in Chinese medicine group and Western medicine group after treatment were decreased at fourteenth day and twenty-eighth day(P 〈0.05) ,and the model group had no change after treatment. The TH positive cells in the model group (49. 83 ± 11.74) were significantly decreased compared with the control group ( 112. 34± 14. 46, P 〈 0. 05 ). The TH positive cells of Western medicine group (86. 29 ±12.28 ) and Chinese medicine group(67.45 ± 13.80) were significantly increased compared with the model group, and the difference was statistically significant ( P 〈 0.05 ). Conclusion Self-made control tremor decoction can improve the rotational behavior of rats with Parkinson disease, and participate in the protection of dopaminergic neurons in the substantia nigra of rats, we can verify the curative effect of self-made control tremor decoction on Parkinson disease by the experimental study.
