摘 要:目的:在细胞水平研究土槿皮乙酸(PAB)抗人肝癌细胞(Hep G2)的机制。方法:以4μmol·L-1PAB分别处理12、24、36、48 h后,通过细胞计数法确定PAB的抑制率;通过细胞形态学观察确定PAB是否诱导细胞死亡;划痕实验观察PAB对肝癌细胞转移的影响;荧光染色法检测PAB处理后Hep G2细胞中微管聚集情况;Realtime PCR检测PAB对P53基因转录的影响;Western blot检测相关蛋白表达。结果:PAB时间依赖性地抑制Hep G2细胞的生长,并且诱导Hep G2细胞的死亡;有效降低肝癌细胞的转移;促进细胞骨架蛋白聚集;降低P53基因转录,并调控P53蛋白和P27蛋白表达。结论:PAB对体外培养的Hep G2细胞生长有明显的抑制作用,并且通过调控微管分布来抑制细胞转移。Objective: The aim of this study was to investigate the anti-tumour role of pseudolaric acid B( PAB) on hepatocellular carcinoma Hep G2. Methods: Cell counting analysis confirmed the inhibitory ratio of PAB. The morphological changes were observed by phase contrast microscopy. Scratch test was applied for cell immigration. Phalloidin –Tetramethylrhodamine B isothiocyanate staining was performed for tubulin polymerization. Real time PCR detected the p53 transcription. Western blot was used for protein expression. Results: The inhibitory role of PAB was in time dependent manner. PAB treatment induced the cell death. Cell immigration was inhibited by PAB treatment. Intervening α-tubulin polymerization was visualized after PAB treatment. Real-time PCR test confirmed that PAB treatment inhibited the transcription of p53 gene. And western blot analysis found that PAB regulated the expression of P53 and P27 protein. Conclusion: Therefore PAB had anti-tumor effect on Hep G2 through various mechanisms.
