摘  要:以南天竹茎段为材料,对不同消毒剂、消毒时间和内生菌抑制剂的消毒效果及诱导、增殖和生根培养基进行研究.结果表明:不同消毒剂消毒效果最佳时间为10min;消毒效果排序1g/L HgCl2>50mL/L NaClO2>20mL/L NaClO2>80mL/L NaClO2;适宜的诱导培养基为MS(murashige skoog培养基)+1mg/L 6-BA+0.1mg/L IBA+90mg/L青霉素+1 600mg/L Vc,诱导率达66.67%;MS+0.5mg/L 6-BA+0.05mg/L NAA是最适增殖配方,株高1.67cm,增殖系数2.63;最佳壮苗培养基为MS+0.01mg/L NAA+0.3mg/L 6-BA,株高2.17cm,增殖系数6.27;在微量元素减半的1/2MS+0.2mg/L NAA+0.2mg/L IBA+4g/L活性碳+30g/L蔗糖的液体培养基上生根率可达76.67%.Effects of different disinfectants,disinfection time and endophyte inhibitor,as well as various media for induction,proliferation and rooting were examined using stem segments of Nandina domestica as material.The results showed that 10 min was the best for all disinfectants,with declining disinfection effect in the order of 1g/L HgCl2,50mL/L NaClO2,20mL/L NaClO2 and 80mL/L NaClO2.The optimum medium for induction was MS+1mg/L 6-BA+0.1mg/L IBA+90mg/L penicillin+1 600mg/L Vc,with induction rate reaching 66.67 %.The optimum multiplication formula was MS+0.5mg/L 6-BA+0.05mg/L NAA,on which the plant height averaged at 1.67 cm and proliferation coefficient was as high as2.63.The best shoot growth was observed on the medium of MS+0.01mg/L NAA+0.3mg/L 6-BA,with mean height of 2.17 cm and proliferation coefficient of 6.27.Meanwhile,the highest rooting percentage of76.67 % was observed on the liquid medium of 1/2 MS(macroelements and microelements reduced by half)+0.2mg/L NAA+0.2mg/L IBA+4g/L activated charcoal+30g/L sucrose.
