异种移植后人卵巢组织的病理学变化

摘  要:目的了解异种移植人卵巢组织病理学的变化。方法无菌条件下,随机将冻融后的人卵巢组织细片移植到不同性别的小鼠皮下,按移植后1周、4周、8周、10周的时间分别处死小鼠,收集移植后人卵巢组织,以解冻后移植前的人卵巢组织作对照,所有组织常规脱水包埋制片,HE染色,镜下观察,比较移植后的人卵巢组织片的卵泡存活、生长、凋亡以及间质重构等情况。结果移植1周后,有大量的新生血管满布于人卵巢组织间质并穿插蔓延,无规律性,与人卵巢组织自然生理血管分布有明显的差别;小鼠性别对移植1周的人卵巢组织中原始卵泡无影响。随着移植时间的推移,卵巢组织间质中的血管逐渐减少,与人卵巢组织正常生理血管分布相似;生长卵泡明显增加。结论人卵巢组织可以移植在不同性别小鼠身上并成活;移植后人卵巢组织的闭锁卵泡及生长卵泡都增加,能产生窦卵泡。Objective To investigate the pathological changes of ovarian tissue in different transplanted human tissues. Methods Under aseptic conditions,the freeze- thaw ovarian tissue was transplanted to different gender mice subcutaneously. The rats were killed respectively at different post- transplantation time after 1 week,4 weeks,8 weeks and 10 weeks; the collected transplantations were compared to the freeze- thaw ovarian tissue without transplantation. All the tissues underwent routine dehydration,embedding section,HE staining and observation under microscope. Follicular survival,growth,apoptosis and interstitial remodeling were compared. Results After transplantation 1 week,a lot of neovascularization overspread disorderly in human ovarian tissue,which is obviously different with physiological vascular distribution; gender is not a fact in primordial follicles. With the passage of time,the vascular distributions in transplantations were similar to normal ovary,with the gradually decreased vessels in the stroma,the growth follicles increased significantly. Conclusion Human ovarian tissue can be transplanted in different gender mice and survive. Both atresia follicles and growth follicles of ovary tissue can be increased,and the antral follicles can be produced.
