摘 要:目的利用基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)快速对致泻性大肠埃希菌鞭毛抗原进行分型。方法收集河南省食品安全风险监测网中分离出的致泻性大肠埃希菌84株,使用MALDI-TOF-MS对其鞭毛抗原进行分型,并与血清学分型结果相比较。结果 84株致泻性大肠埃希菌中76株(90.48%)与血清学分型结果一致,8株分型结果不一致,主要集中在H1和H19、H21和H11。结论 MALDI-TOF-MS分型方法与血清学分型方法相比,在诊断敏感性、诊断特异性、分析敏感性、分析特异性、结果判断、完成检测所需时间、检测粗糙型菌株的能力、稳定性和重现性、吞吐量、检测条件等10个方面均具有较大的优势,更加简便、快捷和准确。MALDI-TOF-MS可以作为一个快速、准确和价廉的工具,应用于致泻性大肠埃希菌鞭毛抗原的分型。Objective To rapidly type E. coli H antigen with matrix- assisted laser desorption ionization- time of flight mass spectrometry( MALDI- TOF- MS). Methods A total of 84 diarrheagenic E. coli isolates were collected from Henan province Food Safety Risk Monitoring Net,and H antigens of those isolates were typed by MALDI- TOF- MS and compared with serologic results,respectively. Results Of the 84 analyzed isolates,76( 90. 48%) were consistent with the results of serological typing. 8 were not consistent,the H antigens of that were mainly H1 and H19,H21 and H11. Conclusion MALDI- TOF-MS type method was compared with the serological typing method,which has advantages in diagnostic sensitivity,diagnostic specificity,sensitivity analysis,analysis of specificity,the judgment,required time to complete test,ability of detecting rough type strains,stability and reproducibility,throughput,and testing conditions,and was more convenient,fast and accurate.MALDI- TOF- MS can be used as a rapid,accurate,and inexpensive tool for the typing of E. coli H antigen.
