摘  要:目的评价不同消毒方式对布鲁菌的消毒效果,为建立高等级生物安全实验室及疫苗生产车间中布鲁菌的消毒标准操作规范提供实验证据。方法筛选粗糙型及光滑型布鲁菌作为模式菌。将模式菌的高浓度菌液和低浓度菌液分别经紫外线、有效氯含量为2 000 mg/ml三氯异氰尿酸及75%乙醇,经过15 min、30 min、60 min杀菌处理,将处理后的细菌置于37℃培养5 d,观察其存活情况,进行消毒效果评价。结果紫外线能在15 min内完全杀灭生物安全柜中的高浓度及低浓度布鲁菌。2 000 mg/ml三氯异氰尿酸在15 min内不能完全杀灭较高浓度布鲁菌,但对低浓度布鲁菌的杀灭效果较好。75%乙醇能在15 min内完全杀灭高浓度及低浓度布鲁菌。结论紫外线、75%乙醇对布鲁菌有较好的消毒效果,可用于高等级生物安全实验室及布鲁菌活疫苗生产车间的常规消毒。Objective To evaluate the effect of different disinfection methods on Brucella,so as to offer experimental data for the establishment of a standard operation procedure in high- level biosafety laboratory and manufacturing shop of Brucellosis vaccine. Methods Rough model bacteria and smooth model bacteria were collected as the model bacteria. Model bacteria in high concentration and low concentration were respectively encountered by ultraviolet radiation,2 000 mg/ml trichloroisocyanuric acid and 75% ethanol for disinfection respectively for 15 min,30 min and 60 min. Thus the survival of bacterial after exposure were evaluated. Bacteria after treatment were set at 37 ℃ for culturing in 5 d,observing the survival,and disinfection effect was evaluated. Results Ultraviolet radiation could kill all the bacteria in high concentration and low concentration in a biosafety cabinet in 15 min. 2 000 mg/ml trichloroisocyanuric acid couldn't kill all the bacteria in high concentration in 15 min,but it had good disinfection effect on bacteria in low concentration. 75% ethanol could kill all the bacteria in high concentration and low concentration in 15 min. Conclusion Ultraviolet radiation and 75% ethanol were effective disinfection methods,thus they are recommended to be use in the regular disinfection of high- level biosafety laboratories and manufacturing shop of Brucellosis vaccine.
