摘 要:目的建立同时测定蒲地蓝消炎口服液中绿原酸和咖啡酸含量的反相高效液相色谱法。方法采用反相高效液相色谱法,色谱柱为岛津Inertsil ODS C18柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.04%磷酸水溶液(15∶85,V/V),流速为1.0 ml/min,柱温为35℃,检测波长为327 nm。结果绿原酸与咖啡酸的质量浓度为4.0 g/ml^19.0 g/ml时,与峰面积呈良好的线性关系,线性回归方程分别为y=37 434.0x-45 435.1(r=0.999 6)、y=111 260x-152 136(r=0.999 8)。绿原酸与咖啡酸的平均回收率分别为99.0%(RSD=1.65%)和100.1%(RSD=1.78%)。结论方法简便、快速、准确,可用于蒲地蓝消炎口服液中绿原酸和咖啡酸含量的同时测定。Objective To establish a reversed-phase high performance liquid chromatography( RP-HPLC) method for the determination of chlorogenic acid and caffeic acid in Pudilan antiphlogistic oral liquid simultaneously. Methods The reversed phase HPLC assay was carried out on a Inertsil ODS C18column( 4. 6 mm × 250 mm,5 μm) with a column temperature at35 ℃,the mobile phase was a mixture of acetonitrile-0. 04% phosphoric acid( 15∶85,V / V) with flowing rate of 1. 0 ml / min,the detection wavelength was set at 327 nm. Results When the linear response range of chlorogenic acid and caffeic acid were among 4. 0 μg / ml-19. 0 μg / ml,a good linear relationship with the peak area. The linear regression equation was y = 37 434. 0x-45 435. 1( r = 0. 999 6),y = 111 260x-152 136( r = 0. 999 8). The average recoveries of chlorogenic acid and caffeic acid were 99. 0%( RSD = 1. 65%) and 100. 1%( RSD = 1. 78%). Conclusion The method was simple,rapid and accurate. It is suitable for the determination of chlorogenic acid and caffeic acid in Pudilan antiphlogistic oral liquid simultaneously.
