摘 要:目的:探讨通天草水提物(AEWH)对APP/PS1双转基因小鼠海马的保护作用。方法:雄性小鼠随机分成对照组,模型组,多奈哌齐组(多奈哌齐0.00092 g·kg^(-1)),AEWH高、中、低剂量组(AEWH 2.5、1.25、0.63 g·kg^(-1))。灌胃给予相同体积的药物或生理盐水,每天灌胃1次,持续35 d。采用Morris水迷宫实验观察小鼠的学习记忆能力,检测小鼠脏器指数,采用双抗体夹心法测定海马白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)含量。结果:与模型组比较,AEWH能明显改善APP/PS1双转基因小鼠的学习记忆能力(P<0.05),明显升高脾和胸腺脏器指数(P<0.05),明显降低海马IL-1β和TNF-α含量(P<0.05)。结论:AEWH能改善APP/PS1双转基因小鼠的学习记忆能力及脾和胸腺脏器指数,机制与降低海马IL-1β和TNF-α水平有关。Objective:To investigate the protective effects of aqueous extract from waternut herb(AEWH)on hippocampal in APP/PS1 double transgenic mice. Methods:Male mice were randomly divided into:control group,model group,Donepezil group(Donepezil 0.00092 g·kg^-1),AEWH high-dose group(AEWH 2.5 g·kg^-1),AEWH medial-dose group(AEWH 1.25 g·kg^-1),AEWH low-dose group(AEWH 0.63 g·kg^-1). All the mice were administrated by intragastric with drugs or water in the same voume for 35 days. The Morris water maze was used to observe the learning and memory ability for the mice. After the treatment all animals were sacrificed,calculated the organ coefficients of mice. Biochemical methods were used to determice the content of IL-1β and TNF-α level in the hippocampal. Results:Compared with the model group,AEWH could significantly improve the learning and memory ability of APP/PS1 double transgenic mice(P〈0.05),the organ coefficients of splenic organ and thymus significantly decreased(P〈0.05). AEWH could decrease the content of IL-1β and TNF-α level in the hippocampal obviously(P〈0.05). Conclusion:AEWH could improve the ability of learning and memory and the organ coefficients of splenic organ and thymus of APP/PS1 double transgenic mice. Its mechanism involve in reducing the content of IL-1β and TNF-α level in the hippocampal.
