兔Perthes病模型建立及股骨头p53、Bcl-2表达研究

摘　　要：目的:建立Perthes病动物模型,观察股骨头相关凋亡基因的变化,探讨其在股骨头坏死中的作用。方法:将32只新西兰幼兔随机分为实验组和对照组,透视下经股骨颈给予实验组(A组)右侧股骨头(实验侧)内注射医用TH胶(含显影剂的d-氰基丙烯酸正辛酯)0.5ml,相同方法给予对照组(B组)注射生理盐水0.5ml,于建模后6、8、10、12周分别取实验组和对照组两组右侧股骨头进行病理观察,观察两组股骨头p53和Bcl-2凋亡基因表达变化。结果:建模后6周A组髋关节开始出现关节间隙增宽,出现点状出血,随着实验期的延长变性区并逐步扩大,股骨头骺板p53基因表达与细胞凋亡率呈正相关性,Bcl-2基因表达阳性率与细胞凋亡呈负相关性。B组股骨头未见明显异常。结论:经股骨颈注射医用TH胶建立Perthes病动物模型稳定,无反复,且操作简单;证实p53和Bcl-2与Perthes病发生相关性。Objective：To establish Perthes disease animal model to observe the changes of apoptosis gene that related to the caput femoris.Methods：32 New Zealand rabbit were randomly divided into experimental group and control group,and in the experimental group（group A）,0.5ml medical TH glue was injected into the right side of the caput femoris under perspective through the collum femoris,0.5ml normal saline was injected to the control group with the same method.After the establishment of the model,the right side of the caput femoris in both groups were observed to see the apoptosis gene expression changes of p53 and BCL-2of caput femoris in the 6th,8th,10 th and 12 th week.Results：After six weeks modeling,group A＇s hip joint gap widened,petechial hemorrhage could be seen.As the extension of the experimental phase degeneration area expanded gradually,the p53 gene expression of femoral epiphyseal plate had positive correlation with apoptosis rate,and the positive rate of BCL-2gene expression was negative related to apoptosis.There was no obvious abnormality in the caput femoris of group B.Conclusion：It is stabile and has no reversal to establish the Perthes disease animal model via the injection of medical TH glue perthes through femoral neck,and the operation is simple.The correlation between p53 and BCL-2and Perthes disease is confirmed.
