补骨脂素对去势骨质疏松雌鼠TGF-β的影响

摘　　要：目的:探讨补骨脂素对去势骨质疏松模型大鼠转化生长因子-β(TGF-β)的影响。方法:将雌性SD大鼠72只随机分为假手术组、模型组、戊酸雌二醇阳性对照组、壮骨止痛胶囊组、补骨脂素高剂量组、补骨脂素低剂量组,每组各12只。除假手术组外,其余各组大鼠制备去卵巢骨质疏松症病理模型,术后5d拆线后连续给药13周,每10d称体质量1次,根据体质量调整用药剂量。给药结束后处死大鼠,采用ELISA法检测血清中雌二醇(E2)、TGF-β含量,提取股骨组织mRNA样本进行PCR反应,检测TGF-β基因的表达。结果:与假手术组比较,模型组血清E2水平明显降低(P<0.01),表明造模成功;与模型组比较,假手术组、戊酸雌二醇阳性对照组、壮骨止痛胶囊组、补骨脂素高剂量组血清TGF-β水平及股骨组织TGF-β基因表达均增加,差异均有统计学意义(P<0.05,P<0.01)。结论:补骨脂素可能通过提高血清TGF-β水平、骨组织TGF-β基因表达发挥抗绝经后骨质疏松症作用。Objective： To investigate the effects of psoralen on the transforming growth factor β（ TGF- β） in a rat model of postmenopausal osteoporosis. Methods： Seventy- two female Sprague- Dawley rats were randomly and equally divided into sham surgery group,model group,estradiol valerate（ positive control） group,Zhuanggu Zhitong capsule group,high- dose psoralen group,and low- dose psoralen group. The rat model of osteoporosis was made by ovariectomy in all groups except the sham surgery group. Rats received respective drugs for 13 consecutive weeks after surgical stitches at 5 days after surgery. The dosage was adjusted according to body mass that was measured every 10 days. All rats were sacrificed after treatment. The enzyme- linked immunosorbent assay was used to determine serum levels of estradiol（ E2） and TGF- β. To determine the expression of TGF- β gene,PCR was performed using mRNA extracted from rat femur. Results： Compared with the sham surgery group,the model group had a significantly lower serum level of E2（ P 0. 01）,indicating that the model was successfully established. Compared with the model group,the sham surgery group,the estradiol valerate（ positive control） group,the Zhuanggu Zhitong capsule group,and the high- dose psoralen group had significantly higher serum levels of TGF- β and significantly higher expression of TGF- β gene in rat femur（ P 0. 05; P 0. 01）. Conclusion： Psoralen antagonizes postmenopausal osteoporosis probably by elevating the serum level of TGF- β and the expression of TGF- β gene in bone.
