摘 要:目的:观察不同中药对饮食诱导肥胖(DIO)大鼠肥胖程度及对瘦素抵抗的影响及黄芪的干预作用。方法:Wistar大鼠分别给予基础饲料和高脂高营养饲料13周,按照体质量分为空白对照组、DIO-R组、DIO模型组、DIO西布曲明组、DIO运脾组、DIO升清组、DIO补脾组,分别以予0.9%氯化钠溶液、西布曲明、苍术和厚朴、柴胡和枳实、黄芪灌胃。12周后,测量体质量、身长、脂肪质量,测定瘦素、神经肽Y(NPY)和细胞因子转录负调节因子(SOCS-3)。结果:与空白对照组比较,DIO模型组体质量升高、SOCS-3明显升高(P<0.05,P<0.01),NPY明显降低(P<0.01);与DIO模型组比较,DIO-R组体质量、脂肪系数、血清瘦素、SOCS-3均明显降低(P<0.01);NPY明显升高(P<0.01);DIO西布曲明组体质量、血清瘦素、SOCS-3降低(P<0.05,P<0.01),脂肪瘦素升高(P<0.01);DIO升清组体质量、血清NPY、脂肪瘦素降低(P<0.01,P<0.05),DIO运脾组NPY、SOCS-3降低;DIO黄芪组体质量、脂肪系数、NPY、瘦素、SOCS-3均降低(P<0.05,P<0.01),且DIO黄芪组较DIO升清组及DIO运脾组更显著升高脂肪瘦素、降低SOCS-3含量(P<0.05,P<0.01)。结论:黄芪能够通过改善瘦素抵抗来抑制肥胖。Objective: To observe the influence of different traditional Chinese medicines on obesity degree and leptin resistance of diet-induced obesity(DIO) rats, and the intervention effects of Radix Astragali seu Hedysari. Methods: Wistar rats were respectively fed with the normal and high-fat diet for 13 weeks. According to the body weight, rats were randomly divided into 7 groups as control group, DIO-R group, DIO model group, sibutramine group, Yunpi group, Shengqing group, and Bupi group. Rats in control and model groups received gavage administration with normal saline, and rats in the treatment groups received gavage administration with sibutramine, Rhizoma Atractylodis and Cortex Magnoliae Officinalis, Radix Bupleuri and immature orange fruit, and Radix Astragali seu Hedysari respectively. The body weight, body length and adipose weight were measured, and the levels of leptin, neuropeptide Y(NPY) and suppressor of cytokine signaling 3(SOCS-3) were examined by using ELSIA after 12 weeks. Results: Compared with control group, the body weight and SOCS-3 level of rats in DIO model group increased, and with a statistical significance(P0.05, P0.01), while the NPY level of rats in this group decreased significantly(P0.01). Compared with model group, the body weight, fat coefficient, levels of serum leptin and SOCS-3 of rats in DIO-R group decreased significantly(P0.01), but the level of NPY increased significantly(P0.01). The body weight, levels of serum leptin and SOCS-3 of rats in sibutramine group decreased significantly(P0.05, P0.01), while the levels of adipose leptin increased significantly compared with model group(P0.01). The body weight, levels of serum NPY and adipose leptin of rats in Shengqing group were all decreased significantly compared with model group(P0.01, P0.05); The levels of NPY and SOCS-3 of rats in Yunpi group decreased significantly, and the body weight, levels of NPY, leptin and SOCS-3 of rats in Bupi group were all decreased significantly compared
