摘 要:目的:建立用HPLC法同时测定加味小柴胡提取物中8种成分的含量方法。方法:色谱条件包括色谱柱Kromasil C18(250mm×4.6mm,5μm),流动相为乙腈-0.005%磷酸水溶液,梯度洗脱,流速1m L/min,检测波长210nm,柱温30℃。结果:方中苦杏仁苷、黄芩苷、汉黄芩苷、柴胡皂苷a、柴胡皂苷d、黄芩素、汉黄芩素、千层纸素A 8种成分在60min内得到分离;每种成分在各自的线性范围内线性关系良好,平均回收率均在97.4%-100.8%,RSD在1.29%-3.10%(n=6)。结论:该方法可同时分析8种化合物,简便、准确,为该方治疗慢性胰腺炎的活性物质研究提供依据。Objective: To establish the quantitative analysis method for detecting the 8 components in extract of modified Xiaochaihu Decoction simultaneously by using high performance liquid chromatography(HPLC) method. Methods: The chromatographic conditions were as follows: chromatographic column was Kromasil C18(250mm×4.6mm, 5μm), mobile phase comprised with acetonitrile-0.005%phosphoric acid(gradient elution), the analysis was performed at the flow rate of 1.0m L/min, the detection wavelength was 210 nm, and the column temperature was 30℃. Results: Eight components including amygdalin, baicalin, wogonoside, saikosaponin a, saikosaponin d, baicalein, wogonin and oroxylin A were separated within 60 min, and each component showed a good linear relationship within their respective linear range. The average recovery rate was between 97.4% and 100.8%, and the RSD was within the range of 1.29%-3.10%(n=6). Conclusion: This method is simple and accurate, which could be used to determine the contents of 8 components in modified Xiaochaihu Decoction extract simultaneously. It also provids the basis for studying the active substances in this formula for treating chronic pancreatitis.
