鼻敏感方对变应性鼻炎模型大鼠血清IL-17及IL-10含量的影响

摘　　要：目的:观察鼻敏感方对变应性鼻炎(AR)模型大鼠的治疗效果及对大鼠血清白介素-17(IL-17)、白介素-10(IL-10)含量的影响,探讨鼻敏感方治疗AR的作用机制。方法:48只SD大鼠随机分为空白对照组,模型组,中药大、中、小剂量组,阳性对照组,共6组。以卵清蛋白(OVA)为致敏原建立大鼠AR模型,造模成功后,模型组灌胃0.9%氯化钠溶液,中药各剂量组灌胃不同浓度的鼻敏感方水提剂,阳性对照组灌胃氯雷他定水溶液,观察各组大鼠行为表现,测定IL-17、IL-10水平,观察鼻黏膜病理形态变化。结果:中药大、中、小剂量组及阳性对照组较模型组鼻痒、喷嚏、清涕等症状明显减轻(P<0.05,P<0.01),血清IL-17水平显著升高,IL-10水平显著降低(P<0.01)。除中药小剂量组外,其余3组鼻黏膜炎性反应程度显著减轻(P<0.05,P<0.01)。结论:鼻敏感方能有效缓解AR模型大鼠鼻部症状,对其鼻黏膜病理模型具有较好的疗效,其治疗AR可能的机制之一是通过影响IL-17、IL-10水平从而调节Th17/Treg细胞平衡,并且两者呈显著性相关关系,提示IL-17、IL-10在AR发病机制中有重要作用。Objective： To observe the therapeutic effect of Bimingan Formula on allergic rhinitis（AR） model rats and its influenceon contents of IL-17 and IL-10 in serum, in order to explore the mechanism of Bimingan Formulain treating AR. Methods： Forty-eight SD rats were randomly divided into 6 groups as control group, model group, high, middle and low dosage of TCM groups, and positive control group. The AR rat model was established by using ovalbumin（OVA）. After modeling, rats in model group received gavage administrated with normal saline, rats in TCM groups received gavage administration with different dosage of water extract of Bimingan Formula, and rats in positive control group received gavage administration with water solution of loratadine. The behaviors of rats in each group were observed, the levels of IL-17 and IL-10 were detected, and the pathological changes innasal mucosa of rats were also observed. Results： Compared with model group, the symptoms asrhinocnesmus, sneezing and watery rhinorrhea of rats in TCM groups and positive control group were obviously alleviated, and the nasal mucosal inflammation of rats in these groups was also alleviated（P0.05, P0.01）. Compared with model group, the contents of IL-17 in serum of rats in TCM groups and positive control group increased, and the contents of IL-10 in serum of rats in those groups decreased（P0.01）. Except the low dosage of TCM group, the rest groups inflammatory reaction were obviously decreased（P0.05, P0.01）. Conclusion： Bimingan Formula could effectively relieve the nasal symptoms of AR rats, and its effect on AR pathological model is good. One of the possible mechanisms of Bimingan Formulain treating AR might be adjusting the balance of Th17/Treg in cells by influencing the levels of IL-17 and IL-10, which suggested that IL-17 and IL-10 play an important role in AR pathogenesis.
