摘  要:目的:体外研究药对防风-乌梅抑制肥大细胞脱颗粒的作用及新机制,为该药在临床治疗过敏性疾病提供科学依据。方法:体外采用胰蛋白酶刺激后P815细胞方法建立肥大细胞脱颗粒模型,采用RT-PCR、Western blotting、ELISA等,观察药对防风-乌梅对肥大细胞分泌组胺,PARs、PAR-2表达的影响。结果:药对防风-乌梅在体外可抑制肥大细胞分泌组胺及PARs、PAR-2表达,与细胞模型组比较,差异有统计学意义(P<0.05,P<0.01)。结论:药对防风-乌梅可能通过调节PAR-2表达阻断肥大细胞脱颗粒,抑制肥大细胞"瀑布效应",从而达到抗过敏作用。Objective: To study the new mechanism and effect of medicine suits as divaricate saposhnikovia rootsmoked plum on expression of PAR-2 and blocking mast cell degranulation, in order to provide a scientific basis for clinical application of this medicine suits in the treatment of allergic diseases. Methods: Mast cell degranulation model was established by stimulating P815 cell with trypsin in vitro, and then the expressions of PARs and PAR-2 in mast cells and the levels of histamine affected by divaricate saposhnikovia root-smoked plum were observed by using RT-PCR, Western blotting and ELISA. Results: Medicine suits as divaricate saposhnikovia root-smoked plum could inhibit the expression of PARs and PAR-2, and the secretion of histamine in mast cells significantly in vitro, and there was a statistically significance compared with model group(P0.05, P0.01). Conclusion: Medicine suits as divaricate saposhnikovia root-smoked plum could play a role of anti-anaphylactic action through regulating the expression of PAR-2 to block mast cell degranulation, and suppress the waterfall effect of mast cells.
