上海地区猪场蚊源乙型脑炎病毒MH-7和PD-19的分离鉴定及分子特征分析

摘  要:为了解上海地区猪场蚊媒携带乙型脑炎病毒(JEV)的流行情况,本研究对2014年上海地区猪场采集的4 000只蚊媒样品进行了病毒分离、RT-PCR及间接免疫荧光鉴定。结果表明,分别从三带喙库蚊和中华按蚊中分离到两株JEV,命名为MH-7株和PD-19株。E基因序列分析显示MH-7株属于基因Ⅰ型,PD-19株属于基因Ⅲ型。与减毒活疫苗株SA-14-14-2株E基因核苷酸和编码氨基酸比较结果显示,MH-7株的同源性分别为87.4%和97.2%;PD-19株为98.1%和97.2%。将两株分离株分别接种3日龄乳鼠,均可在72 h之内致乳鼠死亡,表明它们均具有较强的神经毒力。本研究为上海当地制定JEV防制方案提供了实验依据。To investigate the prevalence of Janpanese encephalitis virus (JEV) from mosquitoes in Shanghai pigpens, a total of 4,000 mosquitoes were collected in 2014. Followed by viral isolation, then identified by RT-PCR and indirect immunofluorescence assay. Two isolates of JEV were isolated from Culex tritaeniorhynchus and Anopheles sinensis. Sequence analysis based on the amplified E gene indicated that MH-7 was grouped into genotype I, and PD-19 was classified into genotype III. The homology of the nucleotide and amino acid sequence between MH-7 and vaccine strain SA-14-14-2 was 87.4% and 97.2%; PD-19 and vaccine stain was 98.1% and 97.2%. Moreover, all of the suckling mice died within 72 hrs when inoculated with the isolate. This study provided a basis references for Shanghai local JEV prevention.
