摘  要:目的观察基质细胞源性因子-1α(SDF-1α)/CXC趋化因子受体-4(CXCR4)信号通路对乳腺癌细胞MDA—MB-435侵袭能力的影响。方法将人乳腺癌细胞株MDA—MB-435分为3组:CXCR4小干扰RNA(siRNA)干扰组、阴性对照siRNA组、空白对照组。文时荧光定量聚合酶链反应(FQ—PCR)和Western blot分别检测CXCR4siRNA转染MDA—MB435细胞前后CXCR4mRNA和蛋白表达水平的差异,Transwell侵袭实验榆测CXCR4下调对MDA—MB-435细胞侵袭能力的影响。结果CXCR4siRNA能够显著降低MDA—MB-435细胞中CXCR4mRNA和蛋白水平的表达,差异均有统计学意义(P〈0.05);Transwell侵袭实验中CXCR4siRNA干扰组穿透基底膜的细胞数为(23.4±1.5)个,明显少于阴性对照siRNA组[(66.2±3.0)个]及空白对照组[(68.2±2.2)个],差异均有统计学意义(P〈0.05)。结论CXCR4可能在调节乳腺癌侵袭转移过程中发挥十分重要的作用。Objective To explore the effect of stromal cell - derived factor - 1α ( SDF - 1α)/ CXC chemokine receptor 4 (CXCR4) signaling pathway on the metastasis of MDA - MB - 435 breast cancer cells. Methods MDA - MB - 435 breast cancer cells were divided into three groups : CXCR4 small interfering RNA (siRNA) intervention group, control siRNA group and blank control group. CXCR4 siRNA was transfected into MDA - MB - 435 cells by lipofection. The mRNA and protein expression of CXCR4 was detected by quantitative real -time fluorescent quantitative polymerase chain reaction (FQ -PCR) and Western blotting. The invasiveness was measured by Transwell invasion assay. Results PCR analysis showed that the expression levels of CXCR4 mRNA in the CXCR4 siRNA intervention group was significantly decreased as compared to the control siRNA intervention group and blank control group (0. 45 vs. 0. 95, 1.00 ; all P 〈 0. 05 ). Similar results were obtained for protein expression as revealed by Western blotting analysis. Transwell invasion assay revealed that the number of CXCR4 siRNA cells (23.4 ± 1.5 ) penetrating the basilar membrane was significantly less than that in control siRNA cells (66. 2 ±3.0, P 〈 0. 05 ) and the blank control cells ( 68.2 ±2. 2, P 〈 0. 05 ). Conclusion CXCR4 might be an important factor for the invasion ability of breast cancer cells.
