三七通舒胶囊的人体药动学和生物等效性研究

摘　　要：目的:建立同时测定人血浆中人参皂苷Rg1、三七皂苷R1浓度的HPLC-MS/MS检测法,研究三七通舒胶囊的肠溶微丸制剂和原制剂在人体的药动学并评价其生物等效性。方法:23名健康男性志愿者分别口服三七通舒胶囊肠溶微丸制剂或原制剂,采用HPLC-MS/MS法测定给药后不同时间点血浆中人参皂苷Rg1和三七皂苷R1的浓度,采用Phoenix Win Nonlin 6.3软件计算其药动学参数,评价2种制剂的生物等效性。结果:人参皂苷Rg1试验制剂与参比制剂的Cmax分别为(1.86±0.63)和(1.77±0.62)ng·m L-1;Tmax分别为(2.33±1.31)和(2.46±1.28)h;AUC0-t分别为(11.35±5.46)和(10.59±4.97)ng·h-1·m L-1;AUC0-∞分别为(12.40±5.69)和(11.34±5.32)ng·h-1·m L-1;三七皂苷R1试验制剂与参比制剂的Cmax分别为(0.77±0.24)和(0.74±0.24)ng·m L-1;Tmax分别为(2.23±1.29)和(2.42±1.30)h;AUC0-t分别为(4.15±1.88)和(3.71±1.60)ng·h-1·m L-1;AUC0-∞分别为(4.62±2.12)和(4.11±1.78)ng·h-1·m L-1。结论:三七通舒胶囊肠溶微丸制剂与原制剂具有生物等效性。Objective： To investigate the pharmacokinetic profiles of Sanqi-Tongshu Capsules（ enteric coated pellets） and Sanqi-Tongshu Capsulesand evaluate their bioequivalence. Methods： Sanqi-Tongshu Capsules（ enteric coated pellets） and Sanqi-Tongshu Capsule were given respectively to 23 healthy male volunteers in an open randomized cross-over study. Concentrations of ginsenoside Rg1 and notoginsenoside R1 in plasma were simultaneouslymeasured by avalidated HPLC-MS / MS method. The pharmacokinetic parameters as well as the relative bioavailability were calculatedby Phoenix Win Nonlin 6. 3. Results： The main pharmacokinetic parameters of ginsenoside Rg1 of test formulation and reference formulation were as follows： Cmax（ 1. 86 ± 0. 63） and（ 1. 77 ± 0. 62）ng·mL^- 1; Tmax（ 2. 33 ± 1. 31） and（ 2. 46 ± 1. 28） h; AUC0- t（ 11. 35 ± 5. 46） and（ 10. 59 ± 4. 97） ng·h^- 1·mL^- 1;AUC0- ∞（ 12. 40 ± 5. 69） and（ 11. 34 ± 5. 32） ng·h^- 1·mL^- 1,respectively. The main pharmacokinetic parameters of notoginsenoside R1 of the test formulation and reference formulation were as follows： Cmax（ 0. 77 ± 0. 24） and（ 0. 74 ± 0. 24） ng·mL^- 1; Tmax（ 2. 23 ± 1. 29） and（ 2. 42 ± 1. 30） h; AUC0- t（ 4. 15 ± 1. 88） and（ 3. 71 ± 1. 60）ng·h^- 1·mL^- 1; AUC0- ∞（ 4. 62 ± 2. 12） and（ 4. 11 ± 1. 78） ng·h^- 1·mL^- 1,respectively. Conclusion： The two formulations of Sanqi-Tongshu Capsulesare bioequivalent.
