摘  要:质粒标准分子是指含有外源基因和内源标准基因特异性片段的重组质粒分子。DNA条形码技术是通过对标准目的基因的DNA序列进行分析从而进行物种鉴定的技术。构建基于DNA条形码的质粒标准分子是DNA条形码技术应用于检测实践的要求。本研究将这两种检测鉴定技术相结合应用于检疫性疫霉的检测,构建了11种检疫性疫霉的DNA条形码标准分子,进行了测序验证、均匀性、稳定性和特异性验证。结果表明,构建的质粒标准分子准确度、均匀性、稳定性和特异性均良好,对实际口岸检验检疫工作具有实践应用价值。Plasmid reference molecule is a recombined plasmid containing reference specific fragments of endogenous and exogenous genes. DNA barcoding is a species identification technique by means of reference genes' sequencing and analysis. The construction of plasmid reference molecule based on DNA barcodes could be applied to the requirement of quarantine practice. In this study, these two technologies were combined for the detection and identification of quarantine Phytophthora species. Eleven reference molecules of quarantine Phytophthora DNA barcodes were constructed and tested for sequencing, homogeneity, stability and specificity. The result showed that the characteristics of accuracy, homogeneity, stability and specificity of constructed reference molecules were all very well, and the technologies were practically applicable to port inspection and quarantine.
