摘 要:目的:建立糯稻根药材及其混淆品杂交水稻根红外图谱的鉴别方法,为中药糯稻根的鉴定与品质评价提供方法和依据。方法:采用傅里叶变换红外光谱法(FTIR)对12批次糯稻根药材进行指纹图谱研究,并对其红外图谱数据进行聚类分析,以共有模式建立糯稻根的对照红外指纹图谱进行全谱相似度计算,将糯稻根和水稻根进行红外一维光谱、二阶导数光谱和全谱相似度比较分析,寻找糯稻根及稻根的差异。结果:12批糯稻根的FTIR光谱共有峰明显,相似度均达到95%以上,表明各批次糯稻根组成结构相近,可用于对其进行质量控制;糯稻根和水稻根之间红外一维光谱和二阶导数光谱的相似度不高,说明红外光谱分析具有一定的指纹特征,可以很好地将它们区分。结论:红外光谱法测定快速简便、所需样品量少,所得结果可以为糯稻根的生药鉴定及质量标准的制订提供依据。Objective:To establish an infrared spectroscopy method for distinguishing between Radix Oryzae Glutinosae and Radix Oryzae Sativae,and to provide a new method and basis for the pharmacognostic identification and the quality evaluation of Radix Oryzae Glutinosae accurately.Methods:Fourier transform infrared spectroscopy(FTIR)was used to analyze 12 batches of Radix Oryzae Glutinosae,which were picked up to establish a reference infrared spectra to calculate the similarity between sample's infrared spectra and the reference infrared spectra,and the hierarchical cluster analysis was carried out as well.Based on the infrared spectra of Radix Oryzae Glutinosae and Radix Oryzae Sativae,the one-dimensional infrared spectra,second derivative spectra and the similarity analysis of their infrared spectra were applied to find out the differences between Radix Oryzae Glutinosae and its imitation,respectively.Results:The common peaks in FTIR spectra of 12 batches of Radix Oryzae Glutinosae were obvious and the similarities of FTIR fingerprint of them were all more than 95%,indicating that the composition and structure of Radix Oryzae Glutinosae from different habitats were similar to one another and this method could be used for controlling the quality of Radix Oryzae Glutinosae.Meanwhile,the differences could be found by one-dimensional infrared spectra and second derivative spectra,the similarity of whole FTIR fingerprint was low between Radix Oryzae Glutinosae and its imitation,indicating that the analysis of FTIR spectra had certain characteristics,which could distinguish them effectively.Conclusion:Infrared spectroscopy used in the present work exhibited some advantages on quick procedures and required less samples,which could be served as a scientific basis for the pharmacognostic identification of Radix Oryzae Glutinosae and the establishment of its quality standard.
