摘 要:目的:测定广西壮药鸡骨草总多糖(PACL)、90%醇沉多糖(PACLl)、70%醇沉多糖(PACL2)、40%醇沉多糖(PACL3)等多糖的含量。方法:用传统的水提法提取鸡骨草多糖,并用三氯乙酸法去除蛋白纯化。所得鸡骨草多糖采用分步醇沉法,用90%、70%、40%的乙醇将鸡骨草粗多糖进行分级沉淀,分别得到PACL和PACL1、PACL2、PACL3三个级分。通过苯酚一硫酸法显色,以葡萄糖标准液绘制标准曲线。结果:回归方程为:Y=13.514X+0.0918,线性系数R2=0.9993,鸡骨草各多糖组分PACLl,PACL2,PACL3,PACL的含量分别为19.78%、11.19%,7.72%,4.44%,即各组分含量关系为:PACL1〉PACL2〉PACL3〉PACL。标准品的平均回收率为:98.83%~100.13%,RSD=1.24%。结论:鸡骨草多糖中多糖的含量均较高,高浓度乙醇沉淀得到的鸡骨草多糖含量较高,低浓度乙醇沉淀的鸡骨草多糖的含量较低,而且鸡骨草总糖未经过醇沉的纯化筛选,其多糖的浓度含量相对较小。Objective :To determine the total content of polysaecharide from Guangxi Zhuang Medicine Abrus cantoniensis Hance (PACL), 90 % polysaccharides of alcohol sink(PACL1 ), 70 % polysaccharide of alcohol sink(PACL2), 40 % polysaccharide of alcohol sink(PACL3), etc. Methods: Extract polysaccharide from Guangxi Zhuang Medicine Abrus cantoniensis Hance by traditional water extraction, remove protein purification by trichloroacetic acid, With 90%, 70%, 40% ethanol to fractional precipitate the crude polysaccharide from Abrus cantoniensis Hance respectively by PACL and PACL1, PACL2, PACL3 grade three points. Use Sulfuric acid - phenol method to color development, with standard curve is drawn in the glucose standard solution. Results: Regression equation is: Y = 13. 514 + 0. 0918 X, linear coefficient R2 = 0. 9993, the content of polysaccharide from every component of Abrus cantoniensis Hance PACL1, PACL2, PACL3, PACL are 19.78 %, 11.19 %, 7.72 %, 4.44 % , the relationship between each component content as follows: PACL1 〉 PACL2 〉 PACL3 〉 PACL. The standard of the average recovery is: 98.83% -- 100.13%, RSD = 1. 24 %. Conclusion(s):Abrus cantoniensis Hanee is full of high Content of polysaccharide, high concentration of ethanol to precipitate the content of Abrus cantoniensis Hance' s polysaccharide is higher, low concentration of ethanol to precipitate the content of Abrus eantoniensis Hance' s polysaecharide is lower, in addition, the total content of polysaccbaride from Abrus cantoniensis Hance is not remove purification by alcohol precipitation, the concentration and content of the poiysaccharide are relatively lower.
