益气活血方抗阿霉素心肌毒性的实验研究

摘　　要：目的:探讨益气活血方对阿霉素所致心肌毒性保护作用的机制。方法:Wistar大鼠40只,随机分为A组(空白组)、B组(阿霉素组)、C组(阿霉素+中药低剂量组)、D组(阿霉素+中药高剂量组)。A组予生理盐水2 m L/只隔日灌胃,其余各组均予阿霉素(2.5 mg/kg)隔日腹腔注射,C组与D组同时给予益气活血方(10.5 g/kg;5.25 g/kg)隔日灌胃,检测血清超氧化物歧化酶(SOD)、丙二醛(MDA)、还原型谷胱甘肽(GSH)、乳酸脱氢酶(LDH)含量,并观察心肌组织病理学变化。结果:与A组比较,B组大鼠血清MDA、LDH含量明显增高(P<0.01),SOD、GSH活性明显降低(P<0.01);与B组比较,C组、D组大鼠MDA、LDH含量明显降低(P<0.01),SOD、GSH活性升高(P<0.05或P<0.01)。结论:益气活血方能有效清除氧自由基而发挥抗氧化作用,在保证阿霉素用药剂量前提下有效防治其对心肌的毒性损伤。Objective： To investigate the mechanism of the protective effect of Yiqi Huoxue Prescription on the myocardial toxicity induced by adriamycin. Methods： Forty Wistar rats were randomly divided into A group（ blank group）,B group（ ADM group）,C group（ ADM ＋ low dose group）,D group（ ADM ＋ high dose group）. A group were treated with physiological saline by intragastric administration 2 m L / each. The other groups were treated with adriamycin（ 2. 5 mg /kg） by intraperitoneal injection. Meanwhile,C and D groups were treated with Yiqi Huoxue Prescription（ 10. 5 g / kg;5. 25 g / kg）. Each group were administered once every other day. The contents of SOD,MDA,GSH,LDH in serum were detected and we observed pathological changes of myocardium. Results： Compared with the A group,the contents of MDA and LDH in serum of B group were significantly increased（ P〈0. 01）,SOD and LDH decreased（ P〈0. 01）. Compared with the B group,the contents of LDH and MDA in C and D groups were significantly decreased,SOD and LDH increased（ P〈0. 05 or P〈0. 01）. Conclusion： Yiqi Huoxue Prescription can effectively remove oxygen free radicals and play an antioxidant role,effectively prophylaxis and treatment of the myocardial toxicity under the premise of ensuring the dose of adriamycin.
