摘 要:目的:通过调节左旋聚乳酸(Poly(L-lactic acid),PLLA)/β-磷酸三钙(beta-tricalcium phosphate,β-TCP)复合多孔支架中β-TCP的比例,制备出一种可生物降解、具有一定强度及骨引导性、细胞相容性及生物相容性良好的支架。方法:采用溶剂自扩散-模压成型——盐沥滤、冷冻干燥法制备了β-TCP含量分别为0、10%、20%、30%、40%、50%的PLLA/β-TCP复合多孔支架。用扫面电镜观测不同β-TCP含量的复合多孔支架的微观形貌,用量重法测量其孔隙率,用动静态疲劳试验机检测各组复合多孔支架的力学强度。各组复合多孔支架接种相同量的骨髓间充质干细胞培养3 d后固定,用扫描电镜观察多孔支架爬行细胞的数量以评价骨支架的细胞相容性。用各组骨支架浸提液培养骨髓间充质干细胞,分别于细胞培养第1天、第3天、第5天及第7天规定时间点用CCK8测试盒检测各组细胞的吸光度以评价骨支架的生物相容性。结果:纯左旋聚乳酸为疏水性材料,不利于细胞的粘附生长;PLLA/β-TCP复合多孔支架表面粗糙,β-TCP的引入可以改善支架材料的亲水性及骨引导性,有利于细胞的爬行生长,其中β-TCP含量为30%(P/T30)组支架细胞接种面扫描电镜下观察细胞爬行生长的数量最多;β-TCP的掺入可使复合支架的X线显影良好,可为支架植入体内降解的体外摄片评估提供方便;PLLA/β-TCP复合支架具备良好的细胞相容性及生物相容性。结论:通过溶剂自扩散-模压成型--盐沥滤、冷冻干燥法制备的PLLA/β-TCP复合支架孔隙较丰富、具备一定的强度,细胞相容性及生物相容性良好,有望通过调节其组分而得到一种降解可调的理想的组织工程骨支架。Objective:To develop a biodegradable scaffold with a certain mechanical strength,osteoconduction and good cytocompatibility and biocompatibility by adjusting the proportion of beta-tricalcium phosphate(β-TCP)in Poly(L-lactic acid)(PLLA)/β-TCP composited porous scaffolds. Methods:The PLLA/β-TCP composited porous scaffolds,in which the content of β-TCP was respectively0%,10%,20%,30%,40%,50%,was synthesized by Solvent self diffusion-molding-salt leaching and freeze-drying method. Its microstructure,porosity,mechanical properties,cytocompati-bility and biocompatibility were studied. Results:PLLA is hydrophobic material and its scaffold is bad for cell adhesion. PLLA / β- TCP composited porous scaffold has rough surface;the addition of β- TCP can improve the hydrophilicity and osteoconduction of scaffold,which is good for cell crawl. In all groups,the P/T30 group scaffold had the largest number of crawling cells. The composite scaffold mixingβ-TCP has good X-ray imaging,which can provide convenience for in vitro X ray assessing the degradation of scaffold in vivo.PLLA/β-TCP composited porous scaffolds have reliable cytocompatibility and biocompatibility. Conclusion:The PLLA/β-TCP composited scaffolds fabricated by Solvent self diffusion-molding-salt leaching and freeze-drying method have rich porosity,certain mechanical strength,osteoconduction and good cytoco-mpatibility and biocompatibility. It is hopeful to obtain an ideal tissue engineered bone scaffold with adjustable degradation rate by adjusting the composition proportion.
