摘 要:目的:分析高脂血症与卵巢颗粒细胞的影响,了解对卵巢激素合成功能的改变。方法:高脂喂养大鼠至性成熟期,测定卵巢组织内颗粒细胞的形态改变,免疫组化和Western blot测定性激素受体和生长分化因子9(growth differentiation factor-9,GDF-9)的蛋白水平改变。结果:高脂喂养后,颗粒细胞的出现病理性改变,其雌孕雄激素受体含量减少,GDF-9在卵巢组织内表达减弱,说明高脂喂养后卵巢颗粒细胞功能的减弱。结论:高脂喂养后,颗粒细胞结构出现损伤性改变,激素合成功能下降,与高脂血症下排卵减弱的临床症状相符合,提示高脂血症影响卵巢排卵功能的不良作用可通过损伤颗粒细胞来实现。Objective:To understand the function of ovarian hormone synthesis change through the analysis of the change of hyperlipidemia and granulosa cells. Methods:Rats under high-fat were fed to sexual mature. Ovarian granulosa cells were observed by transmission electron microscope,and sex hormone receptors and growth differentiation factor-9(GDF-9)changes in ovarian granulosa cells were detected by immunohistochemistry and Western blot. Results:After high-fat diet,granule cell pathological changes being found;estrogen,progesterone and androgen receptors levels were reduced obviously;the expression of GDF-9 in ovarian tissues was weakened;ovarian cell function was declined. Conclusion:After high-fat fed,the damage change of granular cell structure and the decline of hormone synthesis function are consistent with the clinical symptoms of weakened ovulation under hyperlipemia,suggesting the role of hyperlipidemia for weaken ovarian ovulation function by injuring granular cell.
