摘 要:目的探讨黄芪多糖和羟基红花黄色素A单用及合用对慢性肾病大鼠肾细胞凋亡的影响。方法采用单侧输尿管梗阻的方法造成大鼠慢性肾病模型。将30只雄性SD大鼠随机分为假手术组、模型组、黄芪多糖(1 g·kg-1)治疗组,羟基红花黄色素A(50 mg·kg-1)治疗组,黄芪多糖(500 mg·kg-1)+羟基红花黄色素A(25 mg·kg-1)治疗组,假手术组和模型组给予等体积0.9%氯化钠溶液。造模后第2日开始给予相应干预措施,连续给药14 d后处死,检测各组大鼠血清肌酐和尿素氮水平,通过HE染色观察肾组织形态变化,通过TUNEL染色法检测肾细胞凋亡,通过蛋白印迹法检测肾组织中凋亡蛋白Bax、Bcl-2和Caspase-3的表达。结果与模型组相比,黄芪多糖和羟基红花黄色素A单用组及合用组都能够降低血清肌酐和尿素氮水平,改善肾组织病理形态和抑制肾细胞凋亡,合用效果优于单用。结论黄芪多糖和羟基红花黄色素A合用发挥肾脏保护作用优于单用,其作用机制可能与抑制肾细胞凋亡有关。Objective To determine the renal protection effect of astragalus polysaccharides (APS) and hydroxysafflor yellow A (HSYA) used alone or in combination on cell apoptosis in rats with chronic kidney disease (CKD). Methods Rat CKD models were prepared by unilateral ureteral obstruction (UUO), and 30 rats were divided into a sham group, a model group, anAPS (1 g·kg-1) group, an HSYA (50 mg·kg-1) group and anAPS (500 mg · kg - 1) + HSYA (25 mg · kg - 1) group. Astragalus polysaccharides and hydroxysafflor yellow A were treated daily from the day after surgery, All rats were sacrificed 14 days after the UUO. The serum creatinine (Scr) and the serum ureanitrogen (BUN) levels were analyzed. The histopathology of kidney tissues was examined by HE staining. Apoptosis of renal cells from the renal tissues was detected by TUNEL staining and the protein expression ofBax, Bcl-2, and Caspase-3 were determined by Western blot. Results Compared with the model rats, the levels of Scr and BUN, the pathological changes and the apoptosis were significantly decreased after treatment with APS, HSYA, and APS + HSYA, with the combination group showing the best effect. Conclusion The APS and HSYA combination may protect against CKD, and the mechanism may be related to its anti-apoptosis effect.
