摘  要:目的揭示谷跗线螨Tarsonemus granarius体内存在泛变应原精氨酸激酶(arginine kinase,AK)。方法从空调滤网灰尘中采集谷跗线螨(约6 000只)并提取总RNA,反转录为cDNA,设计简并引物,PCR克隆谷跗线螨AK片段,采用生物信息学软件分析克隆基因的编码蛋白特性。结果测序和序列分析结果表明,谷跗线螨AK基因的开放阅读框由724个碱基组成,其编码的蛋白相对分子质量约为26 019,等电点为10.10。结论成功克隆了谷跗线螨AK基因,为进一步谷跗线螨AK蛋白的重组表达以及过敏原性分析奠定基础。In order to clarify that the pan-allergen arginine kinase(AK)gene was existed in the body of Tarsonemus granaries,the live mites(about 6 000)were collected from the air conditioning filter,identified as T.granarius.The total RNA was extracted.The cDNA of AK was cloned,using degenerate primers,from the total RNA of T.granarius.The encoded protein characteristics was analyzed by bioinformatics software.The results of sequencing and sequence analysis showed that the full-length open reading frame of cloned cDNA contained 724 bp and the estimated molecular mass was 26 019(pⅠ10.10).It is concluded that AK was successfully obtained,which will be used for the further recombinant expression and allergenic analysis of T.granarius.
