摘 要:试验优选了过山枫(Celastrus aculeatus Merr.)总芪类的提取和纯化工艺条件。以过山枫总芪类提取率为指标,通过正交试验优选提取总芪类的工艺条件;以大孔树脂的比饱和吸附量与总芪类的回收率为指标,通过静态和动态试验优选树脂纯化总芪类的工艺条件。结果表明,最佳提取工艺条件为5倍量95%乙醇加热回流提取1次,每次1 h,总芪类提取率为0.74%。最佳树脂纯化工艺条件为选择DM130树脂,采用20 mm直径的层析柱,上样溶液的总芪类浓度为5.86 mg/m L,10 g树脂上样30 m L,流速控制在3 m L/min,待吸附后先以30%乙醇洗脱200 m L除杂,再用200 m L 50%乙醇解吸。纯化后总芪类的含量达到58.52%,回收率为60.92%。此优选的工艺合理可行,能够较好地从过山枫中提取和纯化总芪类。To optimize the extraction and purification processes of total stilbenoids from Celastrus aculeatus Merr. Orthogonal test was adopted to optimize extraction process by recording the extraction ratio of total stilbenoids as response indexes. Static and dynamic tests were employed to optimize purification process by taking the adsorption capacity of macroporous resin and the recovery of total stilbenoids as index. The optimum extraction process as follow : the total stilbenoids were extracted 1times with 5-fold 95% ethanol for 1 hours at reflux. The extraction ratio of total stilbenoids was 0.74% under these conditions. The optimum adsorption conditions as follows : DM130 macroporous resin as chromatographic material, 20 mm as the diameter of chromatographic column, 30 m L as adsorption volume of sample solution per 10 g resin, 5.86 mg / m L as total stilbenoids concentration in the sample solution, 3 m L/min as flow rate. After adsorption, 200 m L ethanol(30%) were used to wash out the substance, then 200 m L ethanol(50%) were used for desorption. The concentration of total stilbenoids was58.52% after purification, and the recovery of total stilbenoids was 60.92%. The optimum processes were reasonable and feasible. Through these processes, total stilbenoids can be extracted and purified from Celastrus aculeatus Merr. easily.
